玉屏风颗粒2个异黄酮类及4个色原酮类成分的含量测定 被引量:9

HPLC simultaneous determination of two isoflavonoids,four chromones in Yupingfeng granules

摘要 目的:建立同时测定玉屏风颗粒中2个异黄酮类成分(毛蕊异黄酮葡萄糖苷、毛蕊异黄酮)及4个色原酮类成分(升麻素苷、升麻素、5-O-甲基维斯阿米醇苷、亥茅酚苷)含量的高效液相色谱法。方法:采用INERSTIL ODS3(4.6 mm×250 mm,5μm)色谱柱,以乙腈-0.2%磷酸溶液为流动相,梯度洗脱,流速1.0 mL·min-1,检测波长为254 nm。结果:升麻素苷、毛蕊异黄酮葡萄糖苷、升麻素、5-O-甲基维斯阿米醇苷、亥茅酚苷、毛蕊异黄酮线性范围分别为5.3~263.6,2.1~103.6,7.5~376.2,7.4~396.8,2.0~98.8,4.4~220.6μg·mL-1(r≥0.9999);平均回收率(n=6)分别为99.9%,95.1%,95.7%,96.8%,101.7%,99.0%。结论:该法准确、可靠,可用于玉屏风颗粒的质量控制。 Objective: To develop an HPLC method for simultaneous determination of two isoflavonoids, ( calycosin - 7 - O - β - D - glucoside, calyeosin) and four chromones ( prim - O - glucosylcimifugin, cimifugin, 4' - O - β - glucopyranosyl - 5 - O - methylvisamminol, sec - O - glucosylhamaudol) in Yupingfeng granules. Methods: The six components were separated by INERSTIL ODS3 (4. 6 mm × 250 mm,5 μm) column with gradient elution of acetonitrile-0.2% phosphoric acid as the mobile phase, the flow rate was 1.0 mL · min-1, and the detection wavelength was 254 nm. Results:The calibration curves were linear in the ranges of 5.3 -263.6μg mL-1 for prim - O - glucosylcimifugin,2. 1 - 103.6 μg· mL-1 for calycosin -7 - O -β -D - glucoside,7.5 -376. 2 μg mL-1 for cimifugin,7.4 -396. 8μg mL-1 for 4' - O -β - glucopyranosyl - 5 - O - methylvisamminol, 2.0 - 98.8μg ·mL- 1 for sec - O - glueosylhamaudo1,4. 4 - 220. 6 μg · mL - 1 for ealyeosin ( r ≥0. 9999 ). The average recoveries ( n = 6) were 99.9%, 95.1%, 95.7%, 96. 8%, 101.7%, 99.0%, respectively. Conclusion: This method is simple, rapid and accurate with good reproducibility, it is suitable for the quality control of Yupingfeng granules.
