摘要
目的:探讨全反式维甲酸(ATRA)对大鼠肾小球瞬时受体电位阳离子通道6(TRPC6)表达的影响。方法:雄性Wistar大鼠60只,随机分成假手术组、GS组、ATRA组,每组20只,干预12周后处死大鼠。电镜下观察肾小球超微结构改变。免疫组织化学方法检测肾脏组织TRPC6、TGF-β1、Ⅳ型胶原(Col-Ⅳ)的蛋白分布及表达,逆转录-聚合酶链反应(RT-PCR)方法检测肾脏组织TRPC6 mRNA的表达,免疫印迹法(WB)方法检测肾小球TRPC6的蛋白表达。结果 :与GS组相比,ATRA组大鼠GSI显著下降(P<0.01),肾小球TRPC6、TGF-β1、Col-Ⅳ的蛋白表达量显著下降,TRPC6的mRNA表达量显著下降(P<0.05)。结论:ATRA可能通过抑制肾小球TRPC6的表达而减轻GS进展。
Objective To Investigate the effects of all-trans retinoic acid (ATRA) on the expression and of TRPC6 in glomerulus in rats with glomerrulosclerosis(GS). Methods Sixty 8-week-old Wistar male rats were randomly assigned into sham operated group (n = 20), GS group (n = 20), ATRA treatment group (n = 20). All rats were sacrificed at the 12th week. The pathological and ultrastructurechanges were observed under light microscope and transmission electron microscope. The protein expression of TRPC6, TGF-[31 and collagen(Col-IV ) in glomerulus were detected by immunohitochemistry method. The expression of TRPC6 mRNA in glomerulus were detected by reverse transcription-polymerase chain reaction (RT-PCR). The expression of TRPC6 protein in glomerulus were detected by Western blotting (WB). Results The protein expression of TGF-131 and Col-IV in the ATRA treatment group was markedly down-regulated when compared with the GS group. TRPC6 expression (mRNA or protein) in the GS group was elevated compared with that in the SHO group, and ATRA markedly reduced the mRNA and protein expression of TRPC6 compared with the GS group. Conclusions ATRA can reduce the progression of GS through inhibiting the expression of TRPC6.
