摘要
目的观察迷迭香酸对糖尿病大鼠肾脏的保护作用及探讨其可能的作用机制。方法采用链脲佐菌素(STZ)注射法制作糖尿病大鼠模型,随机分为正常组、糖尿病组、迷迭香酸组。灌胃给药8周后,分别测定血糖、24 h尿蛋白排泄率(UAER)、肾小球滤过率(GFR)、肾组织脂质过氧化物(MDA)和超氧化物歧化酶(SOD)及转化生长因子β1(TGF-β1)、Col-Ⅳ、FN的表达。所采用的技术包括生化、分光光度法、放射免疫、免疫组化、光学显微镜及电镜超微结构等。结果迷迭香酸组大鼠的24 h UAER、GFR、肾组织MDA均显著低于糖尿病组(P均<0.01),迷迭香酸组大鼠肾组织SOD活性显著高于糖尿病组(P<0.01)。迷迭香酸组大鼠肾皮质TGF-β1、FN、Col-Ⅳ的表达均显著低于糖尿病组(P均<0.05)。迷迭香酸组大鼠治疗前后血糖比较及与糖尿病组比较均无显著性差异(P均>0.05)。结论迷迭香酸对STZ糖尿病大鼠肾脏有降低UAER、提高肾组织SOD活性、降低肾组织MDA、改善肾小球高滤过状态作用,并可抑制肾皮质TGF-β1表达、抑制细胞外基质增生。其保护机制与改善氧化应激、抗炎及抑制系膜细胞增生有关。
Objective It is to observe the protective effects of rosmarinic acid on the kidney of diabetic rats and to approach the possible mechanism of action.Methods The diabetic rat models were established with streptozotocin(STZ) injection method and were randomly divided into normal group,diabetes group and rosmarinic acid group.After intragastric administration for eight weeks,the blood glucose,24 h Urine protein Excretory rate(UAER),glomerular filtration rate(GFR) and the expression of lipid peroxides(MDA) in nephridial tissue,superoxide dismutase(SOD),transforming growth factor β1(TGF-β1),Col-Ⅳ and FN were detected respectively.The techniques applied included biochemistry,spectrophotography,radio-immunity,immunohistochemisty,light microscope,scanning electron-microscope.Results 24 h UAER,GFR and MDA in nephridial tissue of the rats in rosmarinic acid group were all significantly lowered than that in diabetes group(all P〈0.01) and the activity of SOD in nephridial tissue of the rats inrosmarinic acid group was significantly higher than that in diabetes group(P〈0.01).The expressions of TGF-β1,FN and Col-Ⅳ in renal cortex of the rats in rosmarinic acid group were all significantly lowered than that in diabetes group(all P〈0.05).There were no significant differences on the blood glucose between before and after treatment in rosmarinic acid group and between rosmarinic acid group and diabetes group(all P〉0.05).Conclusion Rosmarinic acid has the actions of lowering UAER,heightening the activity of SOD in nephridial tissue,lowering MDA in nephridial tissue and improving glomerular high filtration state on the kidney of diabetic rats,and can inhibit the expression of TGF-β1 in renal cortex and the hyperplasia of Extracellular Matrix.The protective mechanism is concerned with improving oxidative stress,anti-inflammation and inhibiting the hyperplasia of mesangial cells.
