摘要
目的:研究葛根枳椇子栀子胶囊的解酒效果以及解酒机制。方法:检测葛根枳椇子栀子胶囊对实验小鼠醉酒时间、实验大鼠血醇浓度和尿液排泄的影响,并测定与乙醇代谢过程相关酶的含量和活性。结果:与模型对照组比较,葛根枳椇子栀子胶囊高剂量组小鼠的酗酒潜伏期延长(P<0.05),低、高剂量组小鼠醉酒时间显著缩短(P<0.01);葛根枳椇子栀子胶囊高剂量组大鼠3h血醇浓度及低、中、高剂量组4h血醇浓度均显著低于模型对照组(P<0.05);高剂量组大鼠尿液排泄量较阴性对照组增加(P<0.05);葛根枳椇子栀子胶囊中、高剂量组大鼠肝脏中的P4502E1含量较模型对照组显著升高(P<0.05),低剂量组大鼠肝脏中的ALDH含量较模型对照组显著升高(P<0.05)。结论:葛根枳椇子栀子胶囊能有效延长小鼠醉酒潜伏期、缩短小鼠醉酒时间,有效加速大鼠体内乙醇代谢过程,可能通过提高大鼠体内细胞色素P4502E1和ALDH的含量,促进尿液排泄,从而加快机体内乙醇的代谢。
Objective： To investigate the anti-alcoholism effect of Gegen Zhijuzi Zhizi capsules and related mechanisms of action. Methods： The effect of Gegen Zhijuzi Zhizi capsules on duration of drunkenness in experimental miceand blood alcohol concentration and urine excretion in experimental rats,and the content and activity of enzymes involved in alcohol metabolism were measured. Results： Compared with the model control group,the high-dose Gegen Zhijuzi Zhizi capsule group had a significantly prolonged latency period of alcoholism in mice（ P〈 0. 05）,and the low-and high-dose groups had significantly shortened duration of drunkenness in mice（ P〈 0. 01）. Compared with the model control group,the high-dose Gegen Zhijuzi Zhizi capsule group had a significantly lower blood alcohol concentration at 3 hours,and the low-,medium-,and high-dose groups had a significantly lower blood alcohol concentration at 4 hours（ P 〈0. 05）. The high-dose Gegen Zhijuzi Zhizi capsule group had a significant increase in urine excretion in rats compared with the negative control group（ P 〈0. 05）. Compared with the model control group,the medium-and high-dose Gegen Zhijuzi Zhizi capsule groups had a significant increase in the content of P4502E1 in the liver of rats（ P 〈0. 05） and the low-dose group had a significant increase in the content of aldehyde dehydrogenase（ ALDH） in the liver of rats（ P 〈0. 05）. Conclusion： Gegen Zhijuzi Zhizi capsules can effectively prolong the latency period of alcoholism in mice,shorten the duration of drunkenness in mice,and accelerate alcohol metabolism in rats,probably through increasing the content of cytochrome P4502E1 and ALDH in rats and promoting urine excretion.
