摘要
目的探讨肥胖症患者脂肪组织中的沉默信息调节因子1(SIRT1)m RNA和蛋白表达水平及其在肥胖症治疗中的临床意义。方法选取接受肥胖治疗的患者105例,以体质量指数(BMI)分为肥胖症组(BMI≥23 kg/m2)和对照组(18.5 kg/m2≤BMI<23 kg/m2);取2组患者腹膜脂肪组织。采用实时荧光定量PCR法检测2组患者脂肪组织SIRT1 m RNA表达水平;采用酶联免疫吸附试验(ELISA)检测脂肪组织SIRT1的蛋白表达水平。并分析2组SIRT1蛋白表达水平与BMI、总胆固醇(TC)、三酰甘油(TG)、空腹血糖(FPG)、空腹胰岛素(FINS)以及胰岛素抵抗指数(HOMA-IR)的相关性。结果肥胖症组SIRT1 m RNA和蛋白表达水平均低于对照组(P<0.05)。肥胖症组脂肪组织中TC、TG、FPG、FINS及HOMA-IR均高于对照组(P<0.05)。SIRT1的蛋白表达水平与总TC、TG、FPG、FINS及HOMA-IR呈负相关(r分别为-0.391、-0.941、-0.184、-0.215及-0.990,均P<0.05)。结论肥胖症患者腹膜脂肪组织中SIRT1 m RNA和蛋白表达水平较非肥胖患者均明显下降,且SIRT1蛋白表达水平与肥胖相关指标呈相关性,对临床治疗肥胖症具有一定的指导作用。
Objective To explore expression levels of SIRT1 m RNA and protein in adipose tissue of obese patients,and its clinical significance in the treatment of obesity. Methods A total of 105 patients in our hospital were selected anddivided into obesity group(BMI≥23 kg/m2) and control group(18.5 kg/m2≤BMI<23 kg/m2) according to body mass index(BMI). SIRT1 m RNA expression levels in adipose tissue(collected from peritoneum) were detected by quantitative PCRmethod in two group. SIRT1 protein expression levels of adipose tissue were detected by enzyme-linked immunosorbent(ELISA) in two groups. The relationships of SIRT1 expression, BMI, total cholesterol(TC), triglyceride(TG), fasting plasmaglucose(FPG) and insulin resistance(HOMA- IR) were analyzed between two group. Results SIRT1 m RNA and proteinlevels were significantly lower in obesity group than those of normal group(P<0.05). The values of TC, TG, FPG, FINS andHOMA-IRinadiposetissueweresignificantlyhigherinobesitygroupthanthoseofnormalgroup(P<0.05).Theproteinexpressionlevel of SIRT1 was positively correlated with values of TC, TG, FPG, FINS and HOMA-IR(r=-0.391,-0.941,-0.184, 0.215and-0.990, P<0.05). Conclusion SIRT1 m RNA and protein expression levels in adipose tissue are significantly reducedin obesity patients than those of control group. There is a close relationship between SIRT1 protein level and obesity index,which has certain guidance for clinical treatment of obesity.
