摘要
为了对15份新疆鹅喉羚肌肉DNA样本和138份粪便DNA样本进行扩增,试验利用牛、羊等近缘物种的20对微卫星引物,通过毛细管电泳法筛选适合新疆鹅喉羚的微卫星位点。结果表明:其中10个位点均为高度多态性位点[多态信息含量(PIC)〉0.5)],共检测到74个等位基因片段,平均每个位点7.4个等位基因。PIC范围为0.509—0.827,个体识别率为0.529~0.845,累计个体识别率为0.999。
To amplify 15 DNA samples from muscle of C, azella subgutturosa and 138 DNA samplesfrom feces of Gazella subgutturosa,20 pairs of DNA primers of microsatellites from Bos Taurus, Ovis aries and other closely related species were used to screen suitable microsatellite loci for Gazella subgutturosa by capillary electmphoresis. The results showed that of which 10 loci were highly polymorphicloci, and the polymorphic in- formation content (PIC) was greater than 0.5. A total of 74 allelic fragments were detected with an average of 7.4 alleles per locus. The PIC ranged from 0. 509 to 0. 827, and the power of discrimination ranged from 0. 529 to 0. 845, and the cumulative power of discrimination was 0. 999.
