摘要 为了研究建立能够进行牛个体识别和亲子鉴定的多态性DNA遗传标记系统,我们筛选出STR基因座,采用荧光标记PCR引物,根据各个基因座的片段大小和扩增条件,优化扩增反应体系和热循环参数,研究建立了6个牛STR基因座组成STR复合扩增系统,通过对无关个体的调查,获得各个遗传标记的多态性遗传基础数据,STR基因座的扩增片段长度在80-280bp范围,每个基因座的等位基因数在8个以上,除基因座ETH225的个体识别率为0.693外,其余5个基因座的识别率在0.8以上,具有较高的个体识别率,6个基因座的联合识别率为大于0.9999999。每个基因座的非父排除概率为0.235-0.739,具有较高的非父排除率,6个基因座的联合非父排除率为0.99225686393,通过家系分析,未发现突变现象,符合孟德尔遗传定律,可以进行个体同一认定和亲子鉴定。 To develope polymorphic DNA genetic marker systems which can be applied in routine individual identification and paternity test for cattle , 16 short tandem repeat loci were selected and multiplex amplified as three groups with fluorescence labeling primers according their fragment size and amplification parameters. The polymorphic data were obtained through screening random selected cattle individuals at above loci. The results showed that the size of amplified fragments for all 6 loci was 80 to 280bp. More than eight alleles at each locus were observed .The power of discrimination was higher than 0.8 except the ETH225 locus as 0.683 .The combined power of discrimination was as high as 0.999999 .The Excluding probability of paternity( PE ) for each locus was around 0.235 -0. 739 .The combined PE of 6 loci was 0. 99225686393. In conclusion, the established multiplex containing 1 cattle STR markers with high PD and PE will be helpful in practice .
