摘要
目的:建立一种灵敏的检测内源表达苦味受体活化的方法,为研究苦味中药的化学及生物物质基础提供依据。方法:利用基于微电子阻抗的实时无标记系统监测激活内源表达T2R38受体的方法。结果:T2R38特异性激动剂在NCI-H716,MDA-MB-231以及Hu Tu-80细胞上均呈现良好的剂量依赖关系。结论:基于微电阻的方法实现了对内源表达受体的监测,可以将其应用于基于T2R38受体的中药药性成分研究,为探究中药苦味药性化学和生物基础提供数据支持。
Objective: To develop a sensitive method to detect the expression of endogenous bitter receptor activation, expecting to provide the chemical and biological material reference for further study of bitter taste Chinese herbal medicine. Methods: The real-time marking system based on microelectronics impedance was used to detect the activation of endogenous expression of T2R38 receptor. Results: Positive specific agonist phenylthiocarbamide(PTC) showed a dose-dependent trend in three endogenous cell lines including NCI-H716, MDA-MB-231 and Hu Tu-80. Conclusion: This method realized the operation of monitoring the expression of endogenous receptor with dependence of the micro resistance. It can be applied to study medicinal ingredients of traditional Chinese medicine based on T2R38 receptor research, with the result that it will provide a data support to explore chemical and biological basis of Chinese medicine with bitter flavor.
