职业接触六价铬对人群线粒体MT-TF基因甲基化水平的影响 被引量:2

Mitochondrial MT-TF gene DNA methylation in workers with occupational exposure to hexavalent chromium

摘要 目的探讨职业接触六价铬对工人线粒体DNA中MT-TF基因甲基化水平的影响。方法采用水浴酸消化一电感耦合等离子质谱法检测人群血铬水平,采用胞质阻滞双核微核法检测人群微核率,采用Mass ARRAY分子量阵列技术检测MT-TF基因甲基化水平。结果 57名手工电镀铬工人为暴露组人群的血铬水平为(10.21±1.35)mg/L,明显高于88名无职业接触史工人为对照组的(3.57±0.43)mg/L,差异有统计学意义(P<0.05);暴露组人群双核微核率为(3.23±0.35)‰,明显高于对照人群(2.22±0.24)‰,差异具有统计学意义(P<0.05);暴露组人群中MT-TF基因甲基化水平为(4.41±0.17)%,明显低于对照组人群甲基化水平(5.19±0.24)%,差异具有统计学意义(P<0.05);相关分析结果显示,被调查人群血铬水平与MT-TF基因甲基化水平有相关关系(r=-0.169 2,P=0.041 9),随着血铬水平增加,MT-TF基因甲基化水平有降低趋势。结论职业接触六价铬可引起机体遗传损伤和表观遗传损伤,MT-TF基因DNA甲基化水平降低是潜在的职业铬暴露的生物标志。 Objective To determine the effect of occupational exposure to chromate(Cr)on the DNA methylation levelof MT-TF gene in Cr-exposed workers. Methods Serum concentration of chromium was detected by inductively coupledplasma mass spectrometry(ICP-MS),and the micronucleus rate was measured by cytokinesis-block micronucleus(CBMN)test. Sequenom Mass ARRAY platform was applied to inspect the DNA methylation level of mitochondrial and nuclear DNA.Results When compared with the control group,the Cr- exposed group had significantly higher serum chromium ionconcentration [(10.21±1.35)vs(3.57±0.43)mg/L,P〈0.05)],higher micronucleus rate [(3.23±0.35)‰ vs(2.22±0.24)‰,P〈0.05],but lower DNA methylation level of MT- TF gene [(4.41 ± 0.17)% vs(5.19 ± 0.24)%,P〈0.05]. Correlation analysisshowed that the serum Cr ion concentration was correlated with the methylation level of MT-TF gene(r=-0.169 2,P=0.041 9),with the higher the Cr ion concentration the lower methylation level. Conclusion Occupational exposure to chromate inducesboth genetic and epigenetic damage. The decreased methylation level of MT- TF is a potential biomarker of occupationalexposure to Cr.
