摘要
目的探讨小檗碱对糖尿病并发症痛性糖尿病神经病变(PDN)发生过程中背根神经节(DRG)中PPARβ、PAR-2、TRPV4、P2X3、n NOS表达的影响。方法采用高糖及加入小檗碱处理原代培养的DRG神经元,Western blot及Real-time PCR法检测DRG中PPARβ、PAR-2、TRPV4、P2X3、n NOS的mRNA及蛋白表达水平。结果高糖培养组中PPARβ、TRPV4的蛋白和mRNA表达高于对照组(P<0.05),加入小檗碱后,mRNA水平下降;PAR-2、P2X3、n NOS mRNA表达低于对照组(P<0.05),加入小檗碱后,mRNA水平上升;PPARβ、PAR-2、PAR-2、P2X3、n NOS蛋白表达水平升高(P<0.05),加入小檗碱后,蛋白表达水平降低。结论小檗碱能使高糖引起的DRG的PPARβ、PAR-2、TRPV4、P2X3、n NOS的蛋白和mRNA水平的改变趋向恢复至正常,有助于维持神经元正常功能。
Objective To explore the effect of berberine on factors related to painful diabetic neuropathy( PDN) in dorsal root ganglia( DRG),including PPARβ,PAR-2,P2X3,TRPV4 and n NOS. Methods Primary cultured DRG neurons were cultured in the condition of high-glucose combined with berberine,methods of western blot and real-time PCR were used to test mRNA and protein levels of PPARβ,PAR-2,n NOS,TRPV4 and P2X3 in DRG,respectively. Results The mRNA levels of PPARβ and TRPV4 in DRG cultured in high-glucose group were higher than those in the control group( P〈0. 05). After berberine was added,mRNA levels decreased,mRNA levels of PAR-2,P2X3 and n NOS were significantly less than those in the control group( P〈0. 05),and increased with addition of berberine. The expressions of PPARβ,PAR-2,TRPV4,P2X3,n NOS were higher than those in the control group( P〈0. 05),and were reduced after adding berberine. Conclusion Berberine helps the departured protein and mRNA levels of PPARβ,PAR-2,TRPV4,P2X3 and n NOS in DRG cultured in high glucose trend to normal,which contributing to maintain the normal function of neurons.
