The Expression of Transient Receptor Potential Vanilloid 4 and Protein Kinase Cε in Dorsal Root Ganglion of Diabetic Rats

摘要 目的探讨糖尿病不同时期蛋白激酶Cε(PKCε)和瞬时性受体电位通道4(TRPV4)在大鼠DRG神经元中的表达及意义。方法采用经典糖尿病模型,检测不同病程糖尿病大鼠DRG神经元中PKCε和TRPV4的表达。用免疫荧光方法,观察背根神经节(DRG)在痛性糖尿病神经病变(PDN)过程中PKCε和TRPV4阳性细胞的变化。结果 PKCε和TRPV4在正常大鼠的背根神经节中均有表达。在糖尿病大鼠中,PKCε和TRPV4的表达水平都出现先升高后降低的现象。其中,PKCε在糖尿病第4周的表达水平最高,TRPV4在糖尿病第1周的表达水平最高。结论高糖慢性刺激可导致DRG神经元PKCε和TRPV4的表达改变,在短期内有明显的增加,但2者高峰出现的时间不同。TRPV4的增加依赖于PKCε,而后者错后的高峰说明其参与更多的细胞内机制。 Objective To investigate the expression and significance of PKCε and TRPV4 expression in DRG neuron in diabetic rats at different duration. Methods Classic diabetic rat model was established and used in the study . The positive neuron in DRG was determined by immunofluorescence staining and the changes in PDN were evaluated. Results Both PKCε and TRPV4 were expressed in DRG neuron of control rat. With the diabetic state continue,both of the proteins first increased and then decreased. The peak expression of PKCε was ob- served in 4w group (duration of diabetes was 4 weeks),and the other was in lw group. Conclusion Sustained chronic high level glucose induced expression alteration of PKCε and TRPV4 in DRG neuron. It showed obvious increase in short term, but the peaks were obtained at different time points. Our results indicated that the increase of TRPV4 depends on PKCε, and subsequent peak of PKCε suggested that it maybe involve in more intracellular mechanisms.
