摘要
目的:观察羟氯喹联合甲氨蝶呤对类风湿性关节炎大鼠血清白细胞介素-1(IL-1)、白细胞介素-6(IL-6)和肿瘤坏死因子-α(TNF-α)水平变化的影响,探讨羟氯喹治疗类风湿性关节炎的临床效果。方法:建立大鼠类风湿性关节炎模型,将100只大鼠分为正常组、模型组、羟氯喹组、甲氨蝶呤组及羟氯喹联合甲氨蝶呤组(联合组),采用酶联免疫吸附法检测大鼠外周血IL-1、IL-6和TNF-α的水平,并观察大鼠关节肿胀程度。结果:模型组、羟氯喹组、甲氨蝶呤组及联合组大鼠外周血IL-1、IL-6和TNF-α水平均高于正常组,治疗后羟氯喹组、甲氨蝶呤组及联合组大鼠的IL-1、IL-6和TNF-α水平均低于模型组,联合组IL-1、IL-6和TNF-α水平均低于羟氯喹组及甲氨蝶呤组,比较差异均有统计学意义(P<0.05),羟氯喹组及甲氨蝶呤组大鼠IL-1、IL-6和TNF-α水平比较差异均无统计学意义(P>0.05);治疗后与模型组比较,羟氯喹组、甲氨蝶呤组及联合组大鼠的足趾肿胀程度均明显减轻,联合组足趾肿胀程度更低于羟氯喹组及甲氨蝶呤组,比较差异均有统计学意义(P<0.05),羟氯喹组和甲氨蝶呤组大鼠足趾肿胀程度比较差异无统计学意义(P>0.05)。结论:羟氯喹在治疗类风湿性关节炎方面具有较好的抗炎作用,能够有效抑制和降低类风湿性关节炎大鼠外周血中IL-1、IL-6和TNF-α水平,且羟氯喹与甲氨蝶呤联合用药治疗类风湿性关节炎的效果更佳。
Objective: To observe the influence of Hydroxychloroquine combined Methotrexate for rheumatoid arthritis in rats serum IL-1, IL-6 and TNF-α level change, to investigate the effect of Hydroxychloroquine in the treatment with rheumatoid arthritis.Method: Rats model of rheumatoid arthritis were established, 100 rats were divided into the normal group, model group, Hydroxychloroquine group, Methotrexate group and combined methotrexate hydroxychloroquine group ( combination group ), the rats peripheral blood IL-1, IL-6 and TNF-α level were detected by enzyme linked immunosorbent assay, the degree of joint swelling in rats were observed.Result: The model group, Hydroxychloroquine group, Methotrexate group and combination group rat peripheral blood IL-1, IL-6 and TNF-α level were all higher than the normal group, after treatment, the Hydroxychloroquine group, Methotrexate group and combination group rats IL-1, IL-6 and TNF-α levels were all lower than the model group, the differences were statistically significant ( P〈0.05 ), there were no statistically significants in the Hydroxychloroquine group and Methotrexate group of IL-1, IL-6 and TNF-ot (P〉0.05) .Compared with the model group after treatment, the Hydroxychloroquine group, Methotrexate group and combination group rats joint swelling degree were significantly reduced, the combination group joint swelling degree was lower than the Hydroxychloroquine group and Methotrexate group, the differences were statistically significant ( P〈0.05 ), there were no statistically significants in the degree of joint swelling in Hydroxychloroquine group and Methotrexate group ( P〉0.05 ) . Conclusion: Hydroxychloroquine in the treatment with rheumatoid arthritis has good anti-inflammatory effect, it can effectively restrain and reduce rheumatoid arthritis rat peripheral blood IL-1, IL-6 and TNF-α level, the Hydroxychloroquine combined Methotrexate in the treatment with rheumatoid arthritis results are better.
