摘要
目的:研究子痫前期胎盘中STAT4和STAT6表达量与可溶性血管内皮生长因子受体-1(sFlt-1)及非对称性二甲基精氨酸(ADMA)生成及胎盘缺氧的相关性。方法:选择40例子痫前期孕妇作为研究的子痫前期组(PE组),选择同期在我院待产的40例健康孕妇作为研究的对照组,收集血清测定STAT4、STAT6、sFlt-1、ADMA含量,收集胎盘测定STAT4、STAT6、HIF-1α、HSP70表达量,行超声检查测定子宫螺旋动脉的PI、RI、S/D。结果:PE组胎盘及血清中STAT4的表达量显著高于对照组、而STAT6的表达量显著低于对照组;PE组患者胎盘中HSP70、HIF-1α的表达量显著高于对照组且与STAT4呈正相关、与STAT6呈负相关,血清中sFlt-1、ADMA的含量显著高于对照组且与STAT4呈正相关、与STAT6呈负相关,子宫螺旋动脉的PI、RI、S/D显著高于对照组且与STAT4呈正相关、与STAT6呈负相关。结论:子痫前期胎盘中STAT4高表达及STAT6低表达与胎盘缺氧密切相关。
[ABSTRACT]Objective:To study the correlation of STAT4 and STAT6 expression levels in preeclampsia placenta with soluble fms-like tyrosine kinase-1 (sFlt-1)and asymmetric dimethylarginine (ADMA)generation as well as placental hypoxia. Methods:A total of 40 cases of pregnant women with preeclampsia were selected as preeclampsia group (PE group),40 cases of healthy pregnant women scheduled for delivery in our hospital during the same period were selected as control group.Serum was collected to determine STAT4,STAT6,sFlt-1 and ADMA levels,the placenta was collected to determine STAT4, STAT6,HIF-1αand HSP70 expression,and ultrasonography was conducted to measure uterine spiral artery PI,RI and S/D. Results:STAT4 expression levels in placenta and serum of PE group were significantly higher than those of control group while STAT6 expression levels were significantly lower than those of control group;HSP70 and HIF-1αexpression levels in placenta of PE group were significantly higher than those of control group,positively correlated with STAT4 and negatively correlated with STAT6,sFlt-1 and ADMA levels in serum were significantly higher than those of control group,positively correlatednbsp;with STAT4 and negatively correlated with STAT6,and uterine spiral artery PI,RI and S/D were significantly higher than those of control group,positively related with STAT4 and negatively correlated with STAT6.Conclusion:High expression of STAT4 and low expression of STAT6 in preeclampsia placenta are closely related to placental hypoxia.
