摘要
目的研究当归补血汤在冠脉结扎诱导的大鼠心肌缺血模型中的抗氧化活性。方法将75只健康SD大鼠随机分为5组,分别为假手术组,模型组,地尔硫卓(10 mg·kg^(-1))组,当归补血汤(3 g·kg^(-1))组,当归补血汤(6 g·kg^(-1))组,连续给药7天后,行冠脉左前降支结扎术制备大鼠心肌缺血模型,24h后,采用HE染色检测心肌病理学改变情况;测定各组血清中的白介素(IL)-6,肿瘤坏死因子(TNF)-α,超氧化物歧化酶(SOD),丙二醛(MDA),过氧化氢酶(CAT)和谷胱甘肽过氧化物酶(GPx);采用Western Blot检测大鼠心脏组织氧化应激相关蛋白表达水平。结果当归补血汤(3 g·kg-1,6 g·kg^(-1))能显著改善冠脉结扎诱导的心脏病理学改变;明显降低冠脉结扎诱导的心肌缺血大鼠血清TNF-α、IL-6和MDA的含量;提高血清中SOD、CAT和GPx的活性含量;同时,在当归补血汤的干预下,氧化应激相关蛋白的SOD1、CAT和GPx的表达明显上调,而MDA5蛋白表达明显下调。结论当归补血汤对冠脉结扎致大鼠心肌缺血有保护作用,其作用可能与调节氧化应激相关细胞因子含量及蛋白表达水平有关。
Objective The investigation was designed principally to evaluate the effects of antioxidant activity of Dang Gui Bu Xue Tang (DBT) on coronary artery ligation -induced myocardial ischemia (MI). Methods 75 SD rats were randomly assigned to five groups (with 15 in each group) : sham group, MI group, MI + dihiazem (10 mg · kg-1) group, MI + DBT (3 g · kg-1 ) group and MI + DBT (6 g · kg-1 ) group. Dihiazem and DBT prctreatment were carried out for 7 consecutive days, re- spectively. Coronary artery ligation was conduct to establish MI model. After 24 h, HE staining was used to detect myocardial pathological condition. Serum interleukin (IL) - 6, tumor necrosis factor (TNF) - α, superoxide dismutase (SOD) , malondial- dehyde ( MDA), catalase (CAT) and glutathione peroxidase (GPx) were measured. The expressions of oxidative stress related - proteins were determined through Western blot. Results DBT could obviously improve cardiac pathology condition and reduce the contents of IL - 6 and TNF - α, as well as the level of MDA. The activity of SOD, CAT and GPx were also recovered with DBT pretreatment. In addition, the expressions of SOD1, CAT and GPx were up - regulated, while the expression of MDA5 was down -regulated with DBT pretreatment. Conclusion The cardioprotective effect of DBT on coronary artery ligation - induced MI nlight be associated with anti -oxidant property.
