摘要
目的观察当归补血汤对高脂饮食大鼠心肌的保护作用。方法 Wistar大鼠分为正常对照组、高脂饮食组、当归补血汤低剂量组(6g/kg)、中剂量组(12g/kg)、高剂量组(24g/kg)、罗格列酮对照组(4mg/kg),每组10只;每日灌胃一次,正常组给予普通饲料,其余组高脂饲料喂养。12周后处死大鼠取其心脏组织,HE染色光镜下观察心肌结构变化,检测心脏组织丙二醛(MDA)和超氧化物歧化酶(SOD)水平。结果 HE染色发现高脂组大鼠心肌出现脂质空泡化,肌纤维排列紊乱。当归补血汤中、高剂量和罗格列酮对照组能明显逆转心肌脂质空泡化情况;模型组大鼠心脏组织MDA水平较正常对照组明显升高,SOD水平降低(P<0.05),当归补血汤中、高剂量组MDA水平较模型组明显降低,SOD水平升高(P<0.05)。结论当归补血汤可能通过增强心肌抗氧化能力对高脂饮食大鼠心肌起到保护作用。
Objective To investigate the protective effect of danggui Buxue Decoction on myocardium of high-fat diet rats. Methods 60 Wistar rats were randomly divided into 6 groups:control group,high-fat diet group,Danggui Buxue decoction low-dose group(6 g/kg),middle-dose group(12 g/kg),high dose group(24 g/kg) and rosiglitazone group(4 mg/kg).All drugs were given by gavage once a day.In addition to the control group,the other five groups were fed with high-fat diet.After 12 weeks,the rats were sacrificated,the structural changes of myocardium were observed with HE staining under light microcope,and the levels of myocardial melondialdehyde(MDA) and superoxide dismutase(SOD) in heart tissues were detected.Results HE staining showed that lipid vacuolation appeared in the myocardium of rats in high-fat diet group,but it was significantly inhibit in rosiglitazone group,Danggui Buxue decoction middle and high-dose groups.The MDA level was significantly increased and the SOD activity was decreased in high-fat diet group(P<0.05).Compared with high-fat diet group,middle and high-dose Danggui Buxue decoction could obviously reversed the changes of MDA and SOD levels(P<0.05).Conclusion Danggui Buxue decoction has protective effect on myocardium of high-fat diet rats by reducing the oxidative stress level.
