摘要
以辣椒的下胚轴为外植体 ,在MS + 6 BA 2 .0mg/L +IAA 1.0mg/L培养基中诱导出愈伤组织 .用加入秋水仙碱的培养基和液体浸泡两种方法处理愈伤组织 ,进行离体细胞多倍体诱导 .结果表明 :将加入秋水仙碱的培养基法优于液体浸泡法 ,秋水仙碱浓度为 10 0 0mg/L ,处理时间为 4d时 ,其加倍效果最好 ,加倍率达 80 % .染色体数目鉴定表明 :四倍体染色体数目是2n =4x =4 8,而二倍体染色体数目是 2n =2x =2 4 .
The callus obtained from the hypocotyls and cotyledon of Capsicum annuum L.were treated by adding colchicine into the medium and by colchicine soaking.The result shows that the former method was better than the later.The best result for inducing tetraploid was by adding colchicine 1000mg/L into the medium treated for 4 days,the inducing rate was up to 80%.The chromosome number of the tetraploid was 2n=4x=48,while that of diploid was 2n=2x=24.
