摘要
目的观察FK506和雷帕霉素滴眼液对内毒素诱导的大鼠葡萄膜炎的治疗作用。方法12R实验大鼠随机分为空白溶剂组、0.1%FK506组、0.2%雷帕霉素组,每组各4只,注射脂多糖24h开始分别滴空白溶剂、0.1%FK506、0.2%雷帕霉素滴眼液,4次/d裂隙灯观察摄像,并参照Hoekzema的方法记录临床评分,井进行病理学观察,比较用药组与对照组的差异。结果成功建立大鼠葡萄膜炎模型,0.1%FK506组、0.2%雷帕霉素组的眼部症状评分在注射后1-2d、7d与对照组比较差异无统计学意义(P〉0.05);3-6d与对照组比较差异有统计学意义(P〈0.05),病理学观察示O.1%FK506组、O.2%雷帕霉素组炎症细胞浸润明显小于对照组组。结论0.1%FK506、0.2%雷帕霉素滴眼液均可有效抑制SD大鼠葡萄膜炎的发展。
Objective To study the effects of FK506 and Rapamycin eye drops in the treatment of endotoxin-induce uveitis in rats. Methods Twelve Sprague-Dawley(SD) rats were divided into three groups, which were 0.1% FK506 group(n=4), 0.2% rapamycin group(n=4) and the control group(n=4). Atter the injection of LPS for 24 hours, the above groups were given eye drops of 0.1%FK506, 0.2% rapamycin. The rats were observed by slit lamp microscope for 4 times per day and the symptom scores were recorded according to the methods provided by Hoekzema. Then the eyes were sampled for pathological observation.T test was used to analyze the data. Results The model of EIU in rats was successfully established. There were significant differences in the symptoms of EIU between 0.1%FK506 group and the control group, between 0.2% rapamycin group and the control group after the medication for 3-6 days(P〈0.05). However, there were no significant differences after the medication for 1-2 days(P〉0.05). Pathological observation on the three grops showed that inflammatory cell infiltration in the 0.1% FK506 group and 0.2% rapamycin group alleviated. Conclusions Both 0.1% FK506 and 0.2% rapamycin eye drops can suppress the development of EIU in SD rats effectively.
