摘要
目的研究肾移植受者的孕烷X受体(PXR)基因多态性对他克莫司浓度/剂量比的影响。方法记录患者的体重、他克莫司血药浓度等,用PCR产物直接测序法对肾移植受者进行基因分型,比较不同基因型患者间他克莫司的浓度/剂量比。结果 PXR-C24381A基因多态性与他克莫司浓度/剂量比值明显相关;他克莫司浓度/剂量比,在A/A型患者明显低于C/A型和C/C型患者(P<0.05),而PXR-A7635G基因多态性对其比值无影响(P>0.05)。结论要取得相同血药浓度,PXR-A24381A基因型患者要比PXR-C24381A型和PXR-C24381C型服用的剂量更高。
Objective To investigate the effect of PXR gene polymor- phism on the concentration/dose ratios of tacrolimus in kidney transplant patients. Methods Blood samples were collected from kidney transplant patients who received tacrolimus. The PXR genotypes were determined by the polymerase chain reaction (PCR) - direct sequencing. Tacrolimus tough concentration was measured by ELISA. The concentration/dose ratio were collected. Results PXR - C24381A gene polymorphism was significantly related with the concentration/dose ratios of patients. Patients with A/A genotype carrier was significantly lower than those with C/A genotype or C/C genotype ( P 〈 0.05 ). No difference was found among the PXR - A7635G genotypes ( P 〉 0. 05 ). Conclusion PXR - C24381A polymorphism is correlated with the whole blood concentration/ dose ratio in kidney transplant patients. genotype ; gene polymorphism
