摘要
目的建立双波长HPLC法同时测定感冒灵胶囊中对乙酰氨基酚、马来酸氯苯那敏、咖啡因3种化学药物和野菊花指标性成分蒙花苷含量的方法。方法采用岛津Inertsil ODS-SP柱(5μm,250mm×4.6mm),流动相为甲醇-水-冰醋酸(26∶23∶1),流速1.0m L·min^(-1),检测波长260nm(对乙酰氨基酚、咖啡因和马来酸氯苯那敏)和334nm(蒙花苷),柱温35℃,进样量20μL。结果对乙酰氨基酚在109.22μg·m L^(-1)~1092.20μg·m L^(-1)范围内呈良好的线性关系(r=0.9999),平均回收率为99.09%,RSD为0.95%;咖啡因在2.05μg·m L^(-1)~20.50μg·m L^(-1)范围内呈良好的线性关系(r=0.9999),平均回收率为100.17%,RSD为0.85%;马来酸氯苯那敏在2.06μg·m L^(-1)~20.59μg·m L^(-1)范围内呈良好的线性关系(r=1.0000),平均回收率为99.74%,RSD为1.05%;蒙花苷在9.77μg·m L^(-1)~97.69μg·m L^(-1)范围内呈良好的线性关系(r=0.9998),平均回收率为98.22%,RSD为1.48%。结论该方法精密度、准确度、重复性和耐用性均较好,可作为感冒灵胶囊中4种成分的含量测定方法。
OBJECTIVE To develop a dual-wavelength HPLC method for simultaneous determining three chem-ical drugs i.e.paracetamol,chlorphenamine maleate and caffeine ,and index component of wild dendranthema flower i.e.linarin;to establish the mensuration for determining the dissolution of Ganmaoling capsule .METHODS Chro-matographic column:Shibadzu Inertsil ODS-SP column (5μm,250mm ×4.6mm),mobile phase:methanol-water-gla-cial acetic acid ( 26∶23∶1 ) , flow rate:1.0 mL· min^ -1 , detection wavelength:260 nm ( paracetamol , caffeine and chlorphenamine maleate ) and 334 nm ( linarin ) , column temperature:35℃, sample size:20μL.RESULTS The content of four components detected by dual-wavelength HPLC:paracetamol in 109.22~1092.20μg· mL-1 showed a good linear relationship ( r=0.9999 ) ,the average recovery rate was 99.09%,RSD was 0.95%;caffeine in 2.05~20.50μg· mL-1 showed a good linear relationship ( r=0.9999 ) , the average recovery rate was 100.17%, RSD was 0.85%;chlorphenamine maleate in 2.06~20.59μg· mL-1 showed a good linear relationship ( r=1.0000 ) , the average recovery rate was 99.74%,RSD was 1.05%;linarin in 9.77~97.69μg· mL -1 showed a good linear re-lationship ( r=0.9998 ) , the average recovery rate was 98.22%, RSD was 1.48%.CONCLUSION The estab-lished dual-wavelength HPLC method ,which can determine paracetamol ,caffeine,chlorphenamine maleate and lina-rin simultaneously , had relatively good precision , accuracy , repeatability and durability , and can be used as assay method for determining contents of the four components in Ganmaoling capsule .
