摘要
为了探讨SLA-DQB基因作为猪抗病育种辅助选择标记的可能性,试验采用PCR-RFLP方法对八眉猪SLA-DQB基因外显子2的多态性及其与仔猪腹泻的相关性进行研究。结果表明:八眉猪SLA-DQB基因外显子2多态性并不丰富,仅检测出3个等位基因和5种基因型,其多态信息含量为0.23,遗传杂合度仅为0.24;八眉猪DQB基因外显子2变异程度比较低。该群体偏离了HardyWeinberg平衡(P<0.01)。仔猪腹泻与性别不相关(P>0.05),与DQB基因外显子2的基因型之间呈现极显著相关(P<0.01),AA和AB基因型腹泻评分均极显著高于AC、BB和CC基因型(P<0.01)。不同性别DQB外显子2的基因型对仔猪腹泻的影响不同。在公猪中,AB基因型个体的腹泻评分高于AA、AC、BB、CC个体(P<0.05);而在母猪中,AA和AB腹泻评分极显著高于AC、BB和CC(P<0.01),可见性别影响了DQB外显子2基因型与腹泻之间的关系。说明八眉猪DQB基因外显子2多态性较低,其基因型对仔猪腹泻有影响,可作为猪抗病育种的选择标记。
To investigate the possibility of SLA - DQB gene as a selection - assisted marker for pig disease - resistant and breeding, the exon 2 polymorphism of SLA - DQB gene and its correlation with piglet diarrhea in Bamei pigs were studied using PCR - RFLP in the test. The results showed that the exon 2 polymorphism of SLA - DQB gene in Bamei pigs was not rich, 3 alleles and 5 genotypes were only detected ; the polymorphic information content was 0.23, and its genetic heterozygosity was only 0.24, and the genetic variation of exon 2 of DQB gene in Bamei pigs was very low. The group deviated from the Hardy - Weinberg equilibrium ( P 〈 0.01 ). The piglet diarrhea was not associated ( P 〉 0.05 ) with the gender of Bamei pigs, while it had highly significant correlation ( P 〈 0.01 ) with the exon 2 genotype of DQB gene. The diar- rhea scores of genotypes AA and AB were significantly higher ( P 〈 0. 01 ) than those of genotypes AC, BB and CC ( P 〈 0. 01 ). The exon 2 genotypes of DQB gene in different genders had different effects in piglet diarrhea, in boars, The diarrhea score of individual with genotype AB was higher (P 〈 0.05 ) than that with genotypes AA, AC, BB, and CC in boars, while in the sow, the diarrhea scores of genotypes AA and AB was extremely significantly higher (P 〈 0.01 ) than those of genotypes AC, BB and CC in sows. Thus, gender influenced the relation- ship between the exon 2 genotype of DQB gene and diarrhea. The results indicate that the exon 2 polymorphism of DQB gene in Bamei pigs is very low, and its genotypes have an effect on piglet diarrhea, which can be used as a selectable marker of pig disease - resistant and breeding.
