摘要 目的建立不同炮制方法延胡索药材的高效液相色谱(HPLC)指纹图谱。方法采用HPLC法,以延胡索乙素为内参照物,色谱柱为Extent-C18柱(250 mm×4.6 mm,5μm),流动相为甲醇(A)-0.1%磷酸溶液(B),三乙胺调pH至6.5,梯度洗脱程序为0~10 min时30%~40%A,10~20 min时40%~45%A,20~60 min时55%~80%A,60~70 min时80%A,流速为1.0 mL/min,检测波长为280 nm,柱温为35℃。结果用所建立的方法对14批延胡索药材进行指纹图谱分析,确定了13个共有峰,建立了延胡索药材的共有模式。在14批延胡索样品中有12批样品的指纹图谱相似度在0.9以上。结论该方法简便、准确、重复性好,可为延胡索药材的质量控制提供参考。 Objective To establish the HPLC fingerprint analysis for the quality control of different processing methods of Cordlis Rhizoma. Methods The separation was performed on an Extent- C_(18) analytical column( 250 mm × 4. 6 mm,5 μm) with the mobile phase consisting of methanol( A) and 0. 1% phosphoric acid( B),adjusted p H to 6. 5 with triethylamine,with gradient elution mode at the flow rate of 1. 0 mL / min. Elution procedure: 0- 10 min,30%- 40% A; 10- 20 min,40%- 45% A; 20- 60 min,55%- 80% A; 60-70 min,80% A. The wavelength of measurement was 280 nm and the column temperature was set at 35 ℃. Results 14 batches of Cordlis Rhizoma were determined under the condition described above. 13 peaks were selected on the chromatographic fingerprints of Cordlis Rhizoma,the mutual model of Cordlis Rhizoma was established and the similarities were calculated. The similarity of 12 samples in all 14 samples were more than 0. 9. Conclustion The method is sample and accuracy with good reproducibility and can be used as a quality control method for Cordlis Rhizoma.
