摘要
目的:检测黑素瘤患者血清中miRNA的表达并评估其生物学意义。方法:通过低通量芯片及实时荧光定量反转录聚合酶链式反应(qRT-PCR)检测黑素瘤患者血清中特异性miRNA的表达,进一步通过聚类分析和风险系数评分评估其特异性及敏感性,并观察miRNA表达与患者生存的关系。结果:共检测出4个黑素瘤特异性表达的血清miRNA,具有良好的灵敏度(83.3%)及特异度(85.0%),其中miR-222的表达与黑素瘤患者的生存呈负相关。结论:本研究检测出的4个黑素瘤血清miRNA可能是该病的特异性生物标志物。
Objective: To identify specific miRNA expression in the serum of melanoma patients and to evaluate its biological significance using a variety of analytic methods. Methods: The expression levels of specific miRNA in the serum of melanoma patients were measured with the low-throughput chips and real-time quantitative reverse transcription-polymerase chain reaction (qRT-PCR). Their specificity and sensitivity were assessed using cluster analysis and risk score. The relationship between miR- NA expression and patients' survival rates was further analyzed. Results: Specific expressions of four miRNAs were detected in the serum of melanoma patients, with high sensitivity(83.3%) and specificity(85.0%). Among these miRNAs, the expression levels of miR-222 were negatively correlated with the survival rate of melanoma patients. Conclusion: Specific expressions of four miR- NAs are identified in the serum of melanoma patient~ These miRNAs could be the specific biomarkers of melanoma.
