摘要
目的观察结肠癌患者癌组织中NIK-IKK结合蛋白(NIBP)、周期蛋白E(Cyclin E)和周期蛋白D1(Cyclin D1)表达变化,并探讨其临床意义。方法收集经外科手术切除及消化内科肠镜活检的部分标本130例份,其中正常结肠黏膜、结肠癌各65例份,患者均未接受放化疗。采用免疫组织化学SP法检测结肠癌组织及正常结肠黏膜组织中NIBP、Cyclin E和Cyclin D1表达,并分析三者阳性细胞表达与结肠癌患者临床病理特征的关系;采用Pearson或Spearman相关检验分析NIBP、Cyclin E和Cyclin D1表达的相关性。结果正常结肠黏膜组织与结肠癌组织中NIBP阳性表达率分别为18.46%(12/65)、60.00%(39/65),Cyclin D1阳性表达率分别为3.08%(2/65)、56.92%(37/65),Cyclin E阳性表达率分别为43.08%(28/65)、89.23%(58/65),两种组织中NIBP、Cyclin D1及Cyclin E阳性表达率比较差异有统计学意义(P均<0.05)。NIBP、Cyclin E和Cyclin D1表达与结肠癌患者肿瘤浸润深度、TNM临床分期、淋巴结转移和远处转移有关(P均<0.05)。结肠癌组织中NIBP与Cyclin E、Cyclin D1表达呈正相关(rs分别为0.43、0.32,P均<0.05),Cyclin E、Cyclin D1表达呈正相关(r_s=0.23,P<0.05)。结论 NIBP、Cyclin E及Cyclin D1在人结肠癌组织中呈高表达,三者可能参与了结肠癌的发生发展。
Objective To observe the expression of NIK-IKK binding protein( NIBP),Cyclin E and Cyclin D1 in the tissues of patients with colon cancer,and to explore its clinical significance. Methods We collected 130 cases of samples from surgical resection and enteroscope biopsy of digestive internal medicine,65 cases of normal colon mucosa and 65 cases of cancer tissues in each group,all patients did not receive radiotherapy and chemotherapy. The expression of NIBP,Cyclin E and Cyclin D1 was detected by immunohistochemistry SP method,and then we analyzed the relationship between the positive expression of proteins and the clinicopathologic features in patients with colon cancer. The correlations between expression of NIBP,Cyclin E and Cyclin D1 were determined by Pearson or Spearman tests. Results The positive expression rates of NIBP in the normal colonic mucosa and colon cancer tissues were 18. 46%( 12/65) and 60. 00%( 39/65),respectively; the positive expression rates of Cyclin D1 were 3. 08%( 2/65) and 56. 92%( 37/65),respectively; and the positive expression rates of Cyclin E were 43. 08%( 28/65) and 89. 23%( 58/65),respectively. Significant differences were found in the positive expression rates of NIBP,Cyclin E and Cyclin D1 between these two groups( all P〈0. 05). The expression of NIBP,Cyclin E and Cyclin D1 was significantly related with the depth of tumor invasion,TNM staging,lymph node metastasis and distant metastasis in patients with colon cancer( all P〈0. 05). In the colon cancer tissues,the expression of NIBP was positively correlated with Cyclin E and Cyclin D1 expression( r = 0. 43,0. 32,all P〈0. 05),and the expression of Cyclin D1 and Cyclin E was positively correlated( r = 0. 23,P〈0. 05). Conclusion NIBP,Cyclin E and Cyclin D1 is highly expressed in the human colon cancer tissues,and they may participate in the occurrence and development of colon cancer tissues.
