摘要
目的观察针刺长强穴对FMR1基因敲除小鼠海马PSD-95表达的影响。方法选取28日龄FMR1基因敲除小鼠共30只,分为长强组、非穴组、模型组,每组10只;长强组选用长强穴,单手进针后,采用平补平泻提插手法行针1min,频率100~160次/min,每日1次,连续治疗6日为1个疗程,疗程间间隔1天,治疗两个疗程;非穴组选用右侧胁下固定非经非穴点,治疗操作同长强组;模型组在同等条件下饲养,不予任何干预。应用Morris水迷宫观察小鼠学习记忆能力,免疫组化法检测小鼠海马区PSD-95蛋白的表达。结果与模型组、非穴组相比,长强组治疗后原平台象限游泳时间/总时间比值明显增高(P〈0.05);跨平台次数明显增高(P〈0.05);海马区PSD-95蛋白表达明显增高(P〈0.05)。结论针刺长强穴能提高FMR1基因敲除小鼠学习记忆和记忆能力,其机制可能与调控海马PSD-95蛋白表达有关。
Objective To observe the effect of acupuncture at Changqiang acupoint on the expresssion of postsynaptic density- 95 in the hippocampus region of the FMR1 knockout mice. Methods A total of thirty 28- days FMR1 knockout mices were randomly divided into model group,anon- acupoint group,Changqiang( GV1) group. The Changqiang group take uniform reinforcing- reducing method after insertion,all the needles will stay in the rats for 1 minutes,frequency of 100- 160 Hz a minute,once a day,1times a day,continuous treatment 6 times for 1 course of treatment,treatment interval between 1 day,two courses of continuous treatment. The same acupuncture manipulation as Changqiang group was used in non- acupoint group where non- acupoint in the right rib was selected. The model group was reared under the same conditions without any intervention. The ratio of swimming time in platform quadrant to total swimming time and the times of stride platform in each group were observed by using Morris water maze,and the PSD- 95 protein expression in hippocampal region was measured by immunohistochemical techniques. Results Compared with the model group and the non- acupoint group,the ratio of swimming distance in platform quadrant to total swimming distance was increased( P 〈0. 05); the times of stride platform were increased( P 〈0. 05); the PSD- 95 protein expression in hippocampal region was increased( P 〈0. 05). Conclusion Acupun ture at Changqiang acupoint can respectively improve learning and memory ability in FMR1 knockout mice and the mechanism of the situation may be related to the improving of PSD-95 expression in the hippocampus region.
