摘要
目的探讨谷氨酰胺(Gln)对慢性阻塞性肺疾病(COPD)患者外周血单个核细胞(PBMC)中p38丝裂原活化蛋白激酶(p38MAPK)活性和白细胞介素-8(IL-8)表达的影响。方法选择32例COPD急性加重期(AECOPD)患者为研究对象,设为AECOPD组,将治疗后转为COPD稳定期(SCOPD)的上述患者设为SCOPD组,收集各组PBMC,分别以Gln进行干预,另选择同期16例健康者为健康对照组。实时荧光定量PCR(RT-PCR)法检测5组PBMC中p38MAPK、IL-8的表达水平。结果 (1)AECOPD和SCOPD的空白对照组中p38MAPK、IL-8的表达均高于健康对照组(分别P<0.01,P<0.05),且急性期高于稳定期(P<0.05)。(2)AECOPD Gln组p38MAPK、IL-8的表达低于AECOPD空白对照组(P<0.01);SCOPD Gln组p38MAPK、IL-8的表达低于SCOPD空白对照组(P<0.05)。结论 Gln可抑制COPD患者炎性细胞中p38MAPK通路的活化,并下调IL-8的表达。
Objective To investigate the effects of glutamine (Gin) on the p38MAPK activity and IL-8 expression in periph- eral blood mononuclear cells(PBMC) of the patients with chronic obstructive pulmonary disease(COPD). Methods Thirty-two pa- tients with acute exacerbation of COPD (AECOPD) were chosen as the research subjects(AECOPD group). Then the patients con- verting to the stable stage of COPD after treatment were set as the SCOPD group. PBMC of each group were extracted and conduc- ted the Gln intervention. At the same time 16 healthy people were selected as the healthy control group. The real-time PCR(RT- PCR) was used to detect the levels of p38MAPK and IL-8 expression in PBMC among the five groups. Results (1)The expressions of p38MAPK and IL-8 in the AECOPD blank control group and the SCOPD blank control group were both higher than those in the healthy control group (P〈0.01 or P〈0. 05),while the expression levels of p38MAPK and IL-8 in patients with AECOPD was higher than those of patients with SCOPD, the difference was statistically significant (P〈 0. 05). (2)The expression levels of p38MAPK and IL-8 in the AECOPD Gin group were lower than those in the AECOPD blank control group (P〈0.01) ;the expres- sion levels of p38MAPK and IL-8 in the SCOPD Gin group were lower than those in the SCOPD blank control group (P〈0.05). Conclusion Gin could inhibit the p38MAPK pathway activation in inflammatory cells of COPD patients and reduce the expression level of IL-8.
