摘要
目的观察急性淋巴细胞白血病(ALL)患儿外周血铁调素25(Hepc-25)mRNA和蛋白表达情况,并探讨其与ALL化疗疗效的关系。方法选取接受MR方案治疗的白血病患儿93例作为观察组,另选取同期20例健康体检儿童作为对照组。采用实时定量聚合酶链反应(RT-PCR)和酶联免疫吸附法(ELISA)测定两组外周血中Hepc-25 mRNA和蛋白表达水平。分析Hepc-25与ALL疗效的关系。结果白血病患儿外周血Hepc-25mRNA及蛋白表达水平均高于对照组,差异具有统计学意义(P﹤0.05);经治疗好转组患儿外周血Hepc-25mRNA及蛋白表达水平较治疗前均下降(P﹤0.05);而治疗无效组患儿Hepc-25 mRNA及蛋白表达水平较治疗前比较,差异无统计学意义(P﹥0.05)。结论 ALL患儿外周血Hepc-25过表达对疾病复发有一定的预测价值,与白血病患儿的化疗疗效有密切关系。
Objective To study the expression of Hepc-25 mRNA and protein in peripheral blood of children with leukemia and to explore the correlation between expression level and efficacy of chemotherapy. Method 93 cases of children with leukemia who were administered with MR regimen were included as study group, while another 20 cases of healthy children were enrolled as control group;peripheral blood was extracted to determine the Hepc-25 mRNA and pro-tein levels using RT-PCR and ELISA; the correlation between Hepc-25 and efficacy was analyzed. Result Hepc-25 mRNA and protein levels in study group were significantly higher than those in the control group (P〈0.05). After treat-ment, Hepc-25 mRNA and protein levels in children with response were significantly decreased (P〈0.05), and there were no significant differences before and after treatment in children without response (P〉0.05). Conclusion The expression of Hepc-25 in peripheral blood of children with leukemia is high, which is useful in predicting recurrence, and is closely correlated with the efficacy of chemotherapy.
