摘要
目的 研究回回甘松饮（HGY）对高糖诱导的肾小管上皮细胞增殖、细胞周期以及Ⅰ型胶原（ColⅠ）和纤维连接蛋白（FN）表达的影响。方法 采用中药血清药理学方法制备含药血清。实验共分为6组：5%空白血清组（NG）、高糖＋5%空白血清组（HG）、高糖＋5%福辛普利钠含药血清组（3 mg/kg）、高糖＋5%HGY含药血清组（20,10,5 g/kg）。细胞以4×10~4个/ml接种于96孔细胞培养板上,分别于药物处理后24,48,72 h时,采用CCK-8法检测NRK-52E增殖情况;细胞以4×10~4个/ml接种于25 cm2细胞培养瓶中,于药物干预48 h时收集细胞,分别采用流式细胞术检测细胞周期变化,免疫酶联吸附法检测ColⅠ和FN蛋白的表达。结果 与NG组比较,高糖（30 mmol/L）能够诱导NRK-52E增殖（P〈0.01）,并能使G_0/G_1期细胞比例减少（P〈0.01）,S期比例增加（P〈0.01）,能够使NRK-52E中ColⅠ和FN蛋白表达量明显升高（P〈0.01）。在上述含药血清干预的24,48,72 h,各含药血清组NRK-52E均受到一定程度抑制,发生G_1/S期阻滞,并可降低高糖条件下NRK-52E中ColⅠ及FN蛋白表达量,与HG组比较有显著差异（P〈0.01或P〈0.05）。结论 HGY含药血清可抑制NRK-52E细胞在高糖环境下的细胞增殖,调控细胞周期,其机制可能与调控ColⅠ和FN蛋白表达有关。
Objective To investigate the effects of HuihuiGansong Yin ( HGY) on the proliferation of rat tubular epithelial cells and its mechanisms.Methods NRK-52E cell lines were divided into 5%blank serum,5%high-glucose+blank serum,5%high-glucose+Fosino-pril sodium drug -containing serum group( 3mg/kg), high-glucose +5%HGY drug c-ontaining serum groups(20,10,5 g/kg).After drug treat-ment for 24,48,72 h,CCK-8 method was used to detect NRK-52E proliferation;flow cytometry was used to detect the cell cycle changes , ELISA was used to test the expressions of FN and ColⅠproteins.Results Compared with 5%blank serum group,high-glucose(30 mmol/L)induced NRK-52E proliferation(P<0.01),and reduced the G0/G1 phase cell proportion(P<0.01),increased S phase proportion (P<0. 01),made the FN and ColⅠexpressions increasing significantly (P<0.01).At the drug-containing serum intervention 24,48,72 h,the G1/S phase retardation,and FN and ColⅠprotein expressions were reduced under the condition of high-glucose(P<0.01 or P<0.05).Conclu-sions HGY drug-containing serum under the high-glucose could make NRK-52E cells in G1/S phase retardation,inhibit the proliferation and its mechanism might be related to regulating FN ,ColⅠprotein expressions .
