摘要
目的为了解吉林省2013年肠道病毒71型(EV71)流行株基因学特征。方法对吉林省2013年手足口病来源的EV71阳性分离物进行VP1编码区逆转录-聚合酶链反应(RT-PCR)扩增,并对扩增产物进行核苷酸序列测定,使用Mega4和Bioedit软件对扩增序列进行基因亲缘关系和氨基酸位点变异分析。结果吉林省2013年分离到的7株EV71同属于C4a基因亚型,并且在VP1编码区第22位氨基酸位点均由谷氨酰胺(Q)突变为组氨酸(H),这一结果与2008年安徽阜阳株突变一致;有2株在VP1编码区第289位点由丙氨酸(A)突变为缬氨酸(V)。结论吉林省2013年的7株EV71流行株与C4a代表株位于同一分支,属于C4a基因亚型,且与2008年阜阳株(EU703813-Fuyang-08/2-C4a)亲缘关系最近,而且发生了氨基酸位点变异。
Objective To understand the gene characteristic of enterovirus 71( EV71) in Jilin province in 2013. Methods The positive isolates of EV71 from hand,foot and mouth disease( HFMD) were conducted for reverse transcription polymerase chain reaction( RT- PCR) amplification on the VP1 coding region in Jilin in 2013,and the amplification products were conducted for nucleotide sequence determination. The amplified sequences was conducted for the analysis of gene phylogeny and amino acid mutation using Mega4 and Bioedit software. Results All of the 7 samples belonged to C4 a subtype,and amino acid sites mutated from Glutamine( Q) to Histidine( H) in the 22 thsite. This result was consistent with the 2008 Anhui Fu yang strain. 2strains of EV71 mutated from Alanine( A) to Valine( V) in the 289 thamino acid site. Conclusion All of the EV71 in Jilin in2013 belongs to C4 a subtype,and has close relationship with the Fuyang strains( EU703813- Fuyang- 08 /2- C4a) in 2008,and the amino acid site variation occurs.
