摘要
目的分析引起吉林省2009年手足口病(Hand Food and Mouth Disease,HFMD)流行的肠道病毒71型(Enterovirus Type 71,EV71)分离株的基因特征。方法选择15株2009年分离于吉林省六个设区的市(州)的HFMD病例的EV71分离株,进行VP1编码区基因全长逆转录-聚合酶链反应扩增、序列测定,使用Bioedit软件和MEGA4.0软件进行同源性、基因亲缘关系分析。结果吉林省2009年的15株EV71分离株均属于C4基因亚型,而且分属于四个相对独立的传播链,其中11株病毒(分离于六个市)属于一个较大的传播链,其余4株形成三个小的传播链。29.5%的吉林省分离株间比对同源性高达99.1%~99.8%,吉林省2009年分离株与安徽省阜阳市2008年以及北京市2008年的流行株间,核苷酸最高同源性分别达到了99.8%和99.7%。结论吉林省2009年至少有四个C4基因型的EV71传播链引起了HFMD的流行,并且有一个EV71优势传播链在几个省以及省内六个市持续传播,引起HFMD的流行;三个小的传播链可能为输入病毒所致。尚未发现在引起HFMD死亡、重症以及轻症的EV71分离株VP1编码区的氨基酸序列呈现规律性的区别。
Objective To analyze the genetic characteristics of enterovirus type 71(EV71)isolates causing hand foot and mouth disease(HFMD)epidemics in Jilin province in 2009. Methods 15 EV71 virus strains from the cases of 6 cities were selected and the RT-PCR and sequences of VP1 genes of EV71 strains were tested and were analyzed by the software of bioedit and Mega4.0. Results All 15 strains of VP1 virus belong to C4 genotype in Jilin province in 2009, which belong to 4 transmission chains respectively. 11 strains of them(isolated from cases of 6 cities)belong to a major transmission chain. The homology among 29.5% provincial virus strains of Jilin province reached 99.1%-99.8%, the highest homology which Jilin isolated strains in 2009 attained 99.8% and 99.7% respectively compared with Fuyang strains in 2008 and Beijing strains in 2008. Conclusion At least 4 transmission chains caused HFMD epidemic in Jilin provinc in 2009, and a dominant transmission chain circulated and caused HFMD epidemic in several provinces and in the 6 cities of Jilin province. Three small transmission chains were caused by imported viruses. There was no the amino differences of the VP1 region among dead, severe and slight HFMD.
