斑蝥酸钠联合化疗方案对非小细胞肺癌患者microRNA-21和SIRT1表达的影响

摘要 目的探讨斑蝥酸钠联合化疗方案对非小细胞肺癌患者微RNA-21(miR-21)和沉默信息调节因子1(SIRT1)表达的影响。方法回顾性分析2008年8月至2014年10月武装警察部队吉林省总队医院收治的87例晚期非小细胞肺癌患者的临床资料,依据治疗方法不同分为研究组(45例)和对照组(42例)。研究组采用斑蝥酸钠联合化疗方案治疗,对照组采用化疗方案。使用实时定量聚合酶链反应法检测miR-21的表达水平,使用免疫组织化学法检测SIRT1的阳性表达率。比较两组患者术前、术后miR-21与SIRT1表达情况。结果治疗后研究组患者的miR-21和SIRT1的表达水平低于对照组[(2.1±1.2)比(2.8±1.4),(40.2±3.1)%比(48.3±3.0)%],差异有统计学意义(P<0.05);研究组患者治疗的总有效率高于对照组[55.6%(25/45)比26.2%(11/42)],差异有统计学意义(P<0.05)。结论使用斑蝥酸钠联合化疗治疗非小细胞肺癌患者可以明显降低患者miR-21与SIRT1的表达,明显改善患者的症状,提高患者的治疗效果,应推广临床应用。 Objective To study the effect of cantharidin sodium combined with chemotherapy on microRNA-21( miR-21) and silent information regulator 1( SIRT1) expression in non-small cell lung cancer patients. Methods Retrospective analysis of clinical data of 87 patients with advanced non-small cell lung cancer in Armed Police Corps Hospital of Jilin Province Force from Aug. 2008 to Oct. 2014 was done,according to the different treatment methods they were divided into a study group( 45 cases) and a control group( 42cases). The study group was treated with sodium antharidate combined with chemotherapy protocols while the control group received only chemotherapy. The expression of miR-21 was detected by real-time PCR and the positive rate of SIRT1 was detected by immunohistochemistry,and the expression of miR-21 and SIRT1 of the two groups before and after therapy were compared. Results After treatment,the expression levels of miR-21 and SIRT1 of the study group were much lower than the control group [( 2. 1 ± 1. 2) vs( 2. 8 ± 1. 4),( 40. 2 ± 3. 1) % vs( 48. 3 ± 3. 0) % ]( P〈0. 05). The total effective rate of the study group was higher than the control group[55. 6%( 25 /45) vs 26. 2%( 11 /42),P〈0. 05]. Conclusion Cantharidin sodium combined with chemotherapy can significantly reduce the expression of miR-21 and SIRT1 in non-small cell lung cancer patients,significantly improve symptoms and the therapeutic effect,thus is worthy of clinical application and promotion.
