摘要
目的 :探讨斑蝥酸钠体外诱导肝癌细胞的凋亡。方法 :应用MTT法、流式细胞仪、透射电镜和琼脂糖凝胶电泳综合分析肝癌细胞的变化。结果 :斑蝥酸钠作用 4 8h使肝癌细胞生长明显抑制 ;细胞凋亡率达 4 0 .0 2 %±2 .0 5 % ,并出现典型的凋亡峰 ,细胞增殖于G2 +M期阻滞 ;细胞核染色质出现凝集、边集等超微结构变化 ;细胞DNA电泳也显示了凋亡特有的“梯子”状条带。结论 :斑蝥酸钠体外可诱导肝癌细胞的凋亡 ,细胞生长阻断在G2 +M期。
Objective: To study sodium cantharidate induced apoptosis of human hepatocellular carcinoma cells in vitro. Methods: The changes of human hepatocellular carcinoma cells treated with sodium cantharidate were analyzed by MTT assay, flow cytometry (FCM), transmission electron microscope (TEM), and agarose electrophoresis assay. Results: Human hepatocellular carcinoma cells were treated with sodium cantharidate for 48h, the cells proliferation was obviously inhibited, the cells showed a typical apoptotic peak and the rate of cells apoptosis was about (40.02±2.05)%, the proliferation of cells was blocked at G 2 +M phase, the chromatin was coagulated nearby the nuclear envelope, and cell DNA electrophoresis showed the typical ladder of apoptosis. Conclusion: Sodium cantharidate can induce human hepatocellular carcinoma cells apoptosis in vitro, and the cells proliferation is blocked at G 2 +M phase.
