马铃薯青枯菌Po82菌株质粒基因组分泌蛋白信号肽的分析 被引量:5

Computational Analysis of Secreting Signal Petides Encoded by Plasmid Genome of Ralstonia solanacearum Po82 Bacteria

摘要 应用计算机技术和生物信息学的方法,采用Signal P4.0、Lipo P1.0、Target P1.0.1、TMHMM2.0蛋白质分析软件对植物病原细菌Ralstonia solanacearum Po82质粒基因组的1680个ORFs编码的分泌蛋白信号肽氨基酸序列进行预测分析。结果表明,质粒基因组的1680个ORFs中有102个ORFs所编码的蛋白质具有N-端信号肽序列,包括85个Ⅰ型信号肽,1个Ⅱ型信号肽,15个Prepilin-like信号肽和1个细菌素和信息素信号肽,所有的分泌蛋白中具有RR-motif信号肽结构的有14个,且均为Ⅰ型信号肽。在102个具有可切割信号肽的分泌蛋白中,有50.98%的蛋白质为胞外分泌型(S型),33.33%为线粒体分泌型(M型),6.87%为叶绿体分泌型(C型),8.82%为其他类型的分泌蛋白。信号肽切割位点的氨基酸组成中非极性氨基酸为53.27%,极性氨基酸为18.79%,带负电荷的酸性氨基酸为19.12%,带正电荷的碱性氨基酸为8.82%。比较R.solanacearum GMI1000和R.solanacearum Po82质粒基因组信号肽类型的区别,发现这2种菌在信号肽类型上存在很大差异。文章通过对R.solanacearum Po82质粒基因组信号肽分泌蛋白的研究,揭示了该病原菌与寄主互作的致病机制在长期进化中的独特地位。 The secreting signal petides amino acid sequences encoded by the 1680 ORFs of the plasmid genome of Ralstonia solanacearum Po82 bacteria were analyzed by using Signal P4. 0,Lipo P1. 0,Target P1. 0. 1,TMHMM2. 0 software. The results showed that the species secreted proteins were 102 ORFs of plasmid genome. The plasmids had 85 ORFs type Ⅰ,1 ORF type Ⅱ,15 ORFs Prepilin-like and 1 ORF Bacteriocins and pheromone signal peptides. In all secreted proteins,the twin-arginine structure was 14 ORFs,and all were type Ⅰ. The102 secreted proteins could be divided into four groups: 50. 98 % for secretory pathway(S),33. 33 % for mitochondrion(M),6. 87 % for chloroplast(C) and 8. 82 % for other types. The frequency of particular amino acids in signal peptides by statistical approach was 53. 27 %for hydrophobic,18. 79 % for uncharged,19. 12 % for negative and 8. 82 % for positive amino acids. Compared with the type of signal peptides between R. solanacearum GMI1000 and R. solanacearum Po82,it revealed that there were significant differences on the two bacteria. This article revealed the unique status about the pathogenic mechanism of the interactions between pathogen and host in a long-term evolution.
