基于Treg-SDF-1/CXCR4-STAT6通路观察阳和平喘颗粒对BMSCs移植后哮喘大鼠气道炎症的影响 被引量:7

Effects of Yanghe Pingchuan Granules on airway inflammation in asthmatic rats after BMSCs transplantation based on Treg-SDF-1/CXCR4-STAT6 pathway

摘要 目的:观察阳和平喘颗粒联合骨髓间充质干细胞(BMSCs)移植对哮喘大鼠Treg-SDF-1/CXCR4-STAT6通路及气道炎症的影响。方法:取50只清洁级雄性SD大鼠,随机分为正常组、模型组、BMSCs组、BMSCs阳和组、BMSCs地米组;采用卵清蛋白致敏激发哮喘,尾静脉移植BMSCs,并予相应药物干预6周。采用HE染色观察肺组织病理形态变化,光镜下计数支气管肺泡灌洗液中各炎症细胞,ELISA检测支气管肺泡灌洗液/血清IL-4、IL-5、IL-13、IgE、IgG1、IgG2a表达,流式细胞术检测外周血CD4+CD2+5Treg、CXCR4,RT-PCR检测肺组织中SDF-1、STAT6 mRNA表达,Western Blot法检测肺组织SDF-1、STAT6蛋白。结果:BMSCs阳和组可见少量黏液分泌,气道上皮细胞增生减轻,气道黏膜及周围组织炎症细胞浸润均减少。与模型组比较,BMSCs阳和组肺泡灌洗液中炎症细胞数量及IL-4、IL-5、IL-13、IgE、IgG1、IgG2a表达降低(P<0.01),外周血CXCR4及肺组织中SDF-1、STAT6 mRNA及蛋白降低,CD4+CD2+5Treg表达升高(P<0.01)。结论:阳和平喘颗粒可调节Treg-SDF-1/CXCR4-STAT6通路,减少气道炎症细胞浸润,从而改善气道炎症。 Objective:To observe the effects of Yanghe Pingchuan Granules combined with bone marrow mesenchymal stem cells(BMSCs)transplantation on Treg-SDF-1/CXCR4-STAT6 pathway and airway inflammation in asthmatic rats.Methods:A total of 50 clean-grade SD rats were randomly divided into normal group,model group,BMSCs group,BMSCs yang group,and BMSCs dimi group.Ovalbumin sensitization was used to induce mice asthma and BMSCs were transplanted into the tail vein.Drug intervention for 6 weeks.HE staining was used to observe the pathological changes of lung tissue.The inflammatory cells in bronchoalveolar lavage fluid were counted under light microscope.Bronchoalveolar lavage fluid/serum IL-4,IL-5,IL-13,IgE,IgG1,IgG2 a were detected by ELISA.Expression of CD4+CD2+5 Treg,CXCR4 were detected by flow cytometry.RT-PCR was used to detect the expressions of SDF-1 and STAT6 mRNA in lung tissue.Western blot was used to detect SDF-1 and STAT6 proteins in lung tissue.Results:A small amount of mucus secretion was observed in the BMSCs yang group,the proliferation of airway epithelial cells was alleviated,and the infiltration of inflammatory cells in the airway mucosa and surrounding tissues was reduced.Compared with the model group,the number of inflammatory cells and the expression of IL-4,IL-5,IL-13,IgE,IgG1,IgG2 a in the BMSCs yang and group alveolar lavage fluid decreased(P<0.01),peripheral blood CXCR4 and lung The expression of SDF-1 and STAT6 mRNA and protein decreased,and the expression of CD4+CD2+5 Treg increased(P<0.01).Conclusion:Yanghe Pingchuan Granules can regulate Treg-SDF-1/CXCR4-STAT6 pathway,reduce airway inflammatory cell infiltration,and improve airway inflammation.
