摘要
目的建立一种稳定高效获得鹿茸片DNA基因组的提取方法,为相似形态中药总DNA提取奠定基础,便于应用分子生物学进行药材鉴定。方法将乌鲁木齐市售鹿茸饮片选取3份为材料,通过两种试剂盒和CTAB三种不同方法提取,提取的DNA再进行琼脂糖凝胶电泳法、紫外分光光度法进行浓度和纯度检测。结果 3种方法均可以从市售鹿茸饮片中提取到基因组DNA,但天根组织提取试剂盒得到的基因组DNA的纯度和浓度最高,提取效果最佳;CTAB法提取效果相对最差。结论天根组织提取试剂盒基因组DNA提取方法操作简便、稳定、高效,提取的DNA效果较好,纯度和浓度检查均合格。
Objective To establish a stable and efficient DNA extraction method of Cornu Cervi Pantotrichum,lay a foundation for similar Traditional Chinese medicine total DNA extraction,be easy to identifying Traditional Chinese medicine by molecular biological technology. Methods Taken 3 Urumqi market Cornu Cervi Pantotrichum as material,the paper extracted the genomic DNA with 3 different methods,including the method of two Genomic DNA kits and CTAB method,and detected the extractive by using the agarose gel electrophoresis and ultraviolet spectrophotometer. Results The results showed that,the DNA extracted by Tiangen Genomic DNA kit was the best,and Easy Pure Genomic DNA kit was better than CTAB,and that extracted by CTAB was the worst. Conclusion The Tiangen Genomic DNA kit was easy,steadily and efficiently to operate,therefore the purity and concentration was qualified.
