摘要
目的通过对5种候选DNA条形码(nrITS,nrITS2,matK,rbcL和trnH-psbA)的筛选找到适合鉴定中国药典山药及其混伪品的DNA序列。方法采用DNA条形码技术,对收集到的山药及其混伪品进行DNA提取,片段扩增和测序。将所得序列在NCBI数据库进行Blast在线比对,确定收到样品集物种正确,用序列比对以及建立系统发育树的方法对山药及其混伪品进行区分。结果通过其成功的扩增和测序,利用DNA序列高种间变异性构建系统发育树,成果鉴定了7个物种样本中5个物种(包括药典规定的山药物种)。结论mat K序列最适合作为薯蓣科薯蓣属药用植物山药鉴定的候选DNA条形码。
Objective In order to find out the DNA sequences whichare suitable for the identification of yam in Chinese Pharmacopoeia and its adulterants by screening five candidate DNA barcodes(nrITS,nrITS2,matK,rbcL and psbA-trnH).Methods DNA barcode technology,DNA extraction,fragment amplification and sequencing were performed on the collected yam and its adulterants.The obtained sequences were blasted through the NCBI database to confirm the correctness of species of the samples.The species of yam and its adulterants were distinguished by sequence alignment and phylogenetic tree.Results Through the successful amplification and sequencing,the phylogenetic trees were constructed with high interspecies variability of DNA sequences.Five species of samples were identified(including the pharmacologically prescribed yam).Conclusion The sequence of mat K is the fittest DNA sequencefor the identification of yam.
