摘要
目的探讨肿瘤-睾丸抗原(NY-ESO-1)、肿瘤蛋白P53(P53)、细胞周期素依赖性蛋白激酶2(CDK2)、膜联蛋白A2(annexin A2)、胰岛素样生长因子2 mRNA结合蛋白1(IMP1)、泛素1(ubiquilin1)以及磷酸甘油酸变位酶(PGAM1)共7种自身抗体在非小细胞肺癌(NSCLC)中独立及联合诊断的应用价值。方法应用Luminex BioPlex200检测48例NSCLC和45例健康对照血清中NY-ESO-1、P53、CDK2、annexin A2、IMP1、ubiquilin1及PGAM1的表达水平,并用受试者工作特征(ROC)曲线分析其诊断的准确性。结果 1)7种自身抗体在NSCLC中的表达均显著高于健康对照组(P<0.05);2)ROC分析显示,NY-ESO-1和P53的AUC均≥0.7,annexin A2的检测特异性为88.89%;3)7种自身抗体联合检测时,AUC为0.812,敏感性为68.75%,特异性为91.11%。结论 7种自身抗体在NSCLC的诊断中均有一定意义,特别是7指标联合检测可有效提高诊断的准确性。
Objective To investigate the diagnostic value of single and combined test of seven serum autoantibodies to tumor related antigens ( NY-ESO- 1, P53, CDK2, annexinA2, IMP1, Ubiquilinl and PGAM1 ) in non-small-cell lung carcinoma. Methods Serum levels of seven autoantibodies in NSCLC patients (48) and health controls (45) were measured simultaneously by multiplexed Luminex assays. Receiving operating characteristic (ROC) curves were used to analyze the diagnostic accuracy in single and combined tests. Results 1 ) The levels of seven autoantibodies in serum were higher in patients with NSCLC than that in health controls ( P 〈 0. 05 ) ; 2 ) The analysis of ROC curves showed that the AUC of NY-ESO- 1 and P53 were more than 0.7, while the sensitivities of annexinA2 was 88.89% ; 3 )The AUC, sensitivity and specificity of combined assay of seven autoantibodies for NSCLC were 0. 812, 68.75% and 91.11%. Conclusions All these seven autoantibodies have certain significance in the diagnosis of NSCLC and combination of seven autoantibodies can enhance the diagnostic accuracy for NSCLC.
