摘要
硫化氢(H_2S)是继一氧化氮(NO)和一氧化碳(CO)之后的第三种内源性气体信号分子,在机体中发挥着重要的生物学效应。研究表明,H_2S参与了心血管系统和神经系统的病理生理过程,并与多种疾病的发生和发展密切相关。本文对近年来H_2S的生物学特性、生理作用、H_2S供体及其相关药物的研究进展做一综述,为相关新药的研发与临床治疗提供有价值的参考。
Hydrogen sulfide(H_2S) is considered as a new member of gasotransmitter family, following nitric oxide(NO) and carbon monoxide(CO). H_2S exerts important biological effects in mammals, which has drawn more and more attention in recent years. It is proved that H_2S has a role in the regulation of physiological and pathophysiological processes in the cardiovascular system and the nervous system. Several cardiovascular and nervous diseases are connected to H_2S. H_2S-releasing agents(also known as H_2S donors) have been widely used not only as useful research tools but also potential therapeutic agents. In this review, we provide an overview of the chemistry and biology of H_2S, and summarize the chemistry and biological activity of some natural and synthetic H_2S donors. We introduce the developments of currently available H_2S-releasing drugs including H_2S-non-steroidal anti-inflammatory drugs, H_2S-nervous system drugs and NO-H_2S-releasing drugs. We hope this review will be a value reference in the development of H_2S-releasing drugs in the treatment of cardiovascular and nervous diseases.
