奥卡西平对戊四氮致癫大鼠脑组织内MMP-2、MMP-9表达的干预作用 被引量:4

摘要 目的:通过戊四氮建立大鼠致癫模型,研究奥卡西平(oxcarbazepine,OXC)对大鼠脑组织内MMP-2与MMP-9表达的干预情况。方法:采用SD雄性大鼠(SPF级),随机分为正常组、戊四氮致癫组以及戊四氮致癫加高中低浓度奥卡西平组。正常组给予生理盐水、戊四氮组给予35mg/kg剂量,戊四氮加奥卡西平组高剂量200mg/kg、中剂量100mg/kg、低剂量组50mg/kg。记录大鼠行为学情况及大鼠疲劳仪上停留时间,应用Real-time PCR方法分析MMP-2、MMP-9 mRNA和蛋白表达量在各组间含量的差异。结果:用戊四氮制备的大鼠癫痫模型,MMP-2与MMP-9表达明显上调,在奥卡西平灌药3周起就能够显著降低MMP-2与MMP-9表达,并且呈现剂量相关性。RT-PCR结果表明正常组的MMP-9与MMP-2的mRNA与蛋白表达水平都处于较低的水平,当注射戊四氮后,MMP-2与MMP-9明显上调(P<0.01,P<0.01),当灌注不同剂量的奥卡西平之后,MMP-9与MMP-2的表达明显下调,200mg/kg奥卡西平组已接近正常组水平。结论:奥卡西平通过对大鼠脑组织内MMP-2、MMP-9表达的干预,对戊四氮致癫大鼠脑组织结构和功能具有保护作用,此保护作用具有剂量相关性。 Objective: To Study MMP- 2 and MMP- 9 expression in the brain tissue by establishing epileptic rat model using oxcarbazepine( oxcarbazepine,OXC). Methods: The subjects with SD male rats( SPF grade)were randomly divided into normal group,pentylenetetrazol pentylenetetrazol epileptic group and low concentration oxcarbazepine group. Normal saline group were given pentylenetetrazole 35 mg / kg dose level. Pentylenetetrazole group were given pentylenetetrazole 35 mg / kg dose level,and concentration oxcarbazepine group were given oxcarbazepine the dose of 200 mg / kg,the dose of 100 mg / kg,the low- dose group 50 mg / kg. The residence time on the case and the fatigue behavior of rats instrument rats were recorded. MMP- 2,MMP- 9 mRNA and protein expression differences in content between the groups were analyzed by real- time PCR. Results: Epileptic rat model was established by using oxcarbazepine( oxcarbazepine,OXC). MMP- 2 and MMP- 9 expression were significantly upregulated in three weeks. Body weight and food intake of rats in each group did not change significantly.MMP- 2,MMP- 9 express after the application of oxcarbazepine treatment were reduced to near normal levels in the control group. The brain structure has been basically removed. RT- PCR results showed that in normal group,MMP- 9 mRNA and protein expression levels of MMP- 2 were at a low level. MMP- 2 and MMP- 9 were significantly increased( P〈0. 01,P〈0. 01). With the injection pentylenetetrazol,MMP- 9 and MMP- 2 expression were significantly reduced. 200 mg / kg oxcarbazepine group was close to normal levels with using different doses of oxcarbazepine. Conclusions: MMP- 2,MMP- 9 expression in the rat brain have a protective effect to brain structure and function by oxcarbazepine intervention with injecting pentylenetetrazol. This protective effect relate to dose.
