摘要
测定杜仲叶中总黄酮和总多糖的含量。以葡萄糖、芦丁为对照品,以蒽酮-硫酸、亚硝酸钠-硝酸铝-氢氧化钠溶液体系为显色剂,采用分光光度法在波长622,506 nm处测定总多糖与总黄酮的含量。结果表明,总多糖的质量浓度在0.004-0.014 mg/m L范围内线性关系良好,回归方程为A=49.0C+0.004(r=0.999 2),总黄酮的质量浓度在0.015 3-0.035 7 mg/m L范围内线性关系良好,回归方程为A=12.0C-0.003 4(r=0.999 7),其平均加标回收率为97.70%,103.01%,RSD(n=6)为3.30%,3.43%。测得总多糖含量为18-36 mg/m L,测得总黄酮含量为52.5-105 mg/m L。分光光度法操作简单、结果准确,具有较好的稳定性和重复性,适用于杜仲叶中总多糖和总黄酮的含量测定;不同时间和不同产地杜仲叶中总多糖和总黄酮的含量存在着差异。
Determination of the content of total flavonoids and total polysaccharides in Eucommia ulmoides oliv. glucose,rutin as reference substance,ketone-sulfuric acid,sodium nitrite-aluminum nitrate-sodium hydroxide solution system as the chromogenic agent,using visible spectrophotometry to detect the content of total polysaccharides and total flavonoids during the wavelength of 622 nm and 506 nm. Results showed that total mass concentration of polysaccharides in 0. 004 - 0. 014 mg / m L range linear relationship is good,the regression equation is A = 49. 0C + 0. 004( r = 0. 999 2),the mass concentration of total flavonoids in 0. 015 3 - 0. 035 7 mg / m L range linear relationship is good,regression equation is A = 12. 0C- 0. 003 4( r = 0. 999 7). Its average recovery was 97. 70%,103. 01%,RSD was 3. 30%,3. 43%( n =6). The content of measured total polysaccharides is 18 - 36 mg / m L. The content of measured total flavonoids is 52. 5 - 105 mg / m L. The method with good stability and repeatability is simple,accurate and suitable for determing the content of total polysaccharides and total flavonoids in leaves of Eucommia ulmoides oliv. The content of total polysaccharides and total flavonoids in leaves of Eucommia ulmoides oliv from different time and different origin has some difference.
