摘要
目的对比分析实时荧光RT-PCR法与常规RT-PCR法在手足口病病原体检测中的差别,并评价其优劣。方法采用实时荧光RT-PCR法与常规RT-PCR法,对手足口病患者的咽拭子标本进行检测,并对检测结果作统计学分析。结果实时荧光RT-PCR法与常规RT-PCR法检测EV71结果差异有统计学意义(χ2=21.62,P<0.01),EV71阳性率分别为76.53%(75/98)和41.84%(41/98),实时荧光RT-PCR法EV71阳性而常规RT-PCR法EV71阴性的样品有7份。实时荧光RT-PCR法与常规RT-PCR法检测Cox A16结果差异有统计学意义(χ2=34.45,P<0.01),Cox A16阳性率分别为13.26%(13/98)和5.10%(5/98),实时荧光RT-PCR法Cox A16阳性而常规RT-PCR法Cox A16阴性的样品有3份。实时荧光RT-PCR法从RNA提取到检测结果需3 h^4 h,常规RT-PCR法需6 h^7 h。结论实时荧光RT-PCR法具有快捷、灵敏、直观等特点,在手足口病病原体检测中具有较大的应用价值。
Objective To compare and analyze the difference between real-time PCR and conventional PCR testing for hand-foot-mouth disease( HFMD) pathogen,and to evaluate the advantages and disadvantages. Methods Throat swab specimens which were collected from patients clinically diagnosed with HFMD,were detected by real-time RT-PCR and conventional RT-PCR. The detection results were analyzed. Results There was statistical significance on the difference in detection of EV71( χ2= 21. 62,P 0. 01),with a respective positive rate for EV71 of 76. 53%( 75 /98) and 41. 84%( 41 /98),real-time RT-PCR detected 7 cases of EV71-positive samples that was negative according conventional RT-PCR. There was statistical significance on the difference in detection of Cox A16( χ2= 34. 45,P 0. 01),with a respective positivity for Cox A16 of13. 26%( 13 /98) and 5. 10%( 5 /98),real-time RT-PCR detected 3 cases of EV71-positive samples that was negative according conventional RT-PCR. The test could be completed in 3 h-4 h from RNA extraction to final results by real-time RT-PCR,and 6 h-7 h by conventional RT-PCR. Conclusion Real-time RT-PCR is rapid,sensitive with visual results and posseses great value for practical application in the detection of HFMD.
