摘要 目的比较实时荧光PCR检测(FQ-PCR)与RT-PCR在鉴定甲型流感病毒中的优劣,建立能快速简便、特异地从临床标本中检测并鉴定流感病毒的新方法。方法对200份临床采集的咽拭子样本进行FQ-PCR与RT-PCR检测,综合比较2种监测方法之间的差异。结果 FQ-PCR检测200份临床咽拭标本,96份甲型阳性,阳性率为48.0%;从RNA提取到检测结果仅需3~3.5 h。RT-PCR检测200份临床咽拭标本,69份阳性,阳性率为34.5%;从RNA提取到检测结果约需8 h。结论 FQ-PCR方法快速、特异、灵敏,在甲型流感病毒检测中具有较大的应用价值。 [Objective]To compare the advantage and disadvantage of real-time fluorescent PCR(GQ-PCR) and RT-PCR in the detection of Influenza A Virus,so as to establish new method for detection and identification of influenza virus from clinical samples simply,rapidly and specially.[Methods]Totally 200 pharyngeal swab samples were tested by FQ-PCR and RT-PCR,the difference between them was compared comprehensively.[Results]96 out of 200 pharyngeal swab samples was positively indentified by FQ-PCR,the positive rate was 48.0%.The test could be completed in 3~3.5 hours from RNA extraction to final results.69 samples were positively indentified by RT-PCR,the positive rate was 34.5%.The test could be completed in 8 hours.[Conclusion] FQ-PCR is rapid,specific and sensitive and possesses great value for practical application in the detection of Influenza A virus.
