miR-21和miR-155在系统性红斑狼疮患者外周血B细胞中的表达及意义 被引量:1

The Expressions and Significances of miR-21 and miR-155 in Peripheral Blood B Cells of Patients with Systemic Lupus Erythematosus

摘要 目的:探讨miR-21和miR-155在系统性红斑狼疮患者外周血B细胞中的表达及意义。方法:将93例系统性红斑狼疮(SLE)患者根据SLEDAL评分分为轻度活动组(n=49)和中重度活动组(n=44),并选择40例健康者作为对照组,采用实时荧光定量PCR技术检测其外周血B细胞中miR-21、miR-155、骨形成蛋白-2(BMP-2)和骨形成蛋白受体Ⅱ(BMPRⅡ)的表达。结果:SLE患者外周血B细胞中mi R-21和mi R-155相对表达量均高于对照组(P<0.05),中重度活动组患者外周血B细胞中miR-21和mi R-155相对表达量均高于轻度活动组和对照组,且轻度活动组高于对照组,差异均有统计学意义(P<0.05);SLE患者外周血B细胞中BMP-2 mRNA和BMPRⅡmRNA相对表达量均高于对照组(P<0.05),中重度活动组患者外周血B细胞中BMP-2 mRNA和BMPRⅡmRNA相对表达量均高于轻度活动组和对照组,且轻度活动组高于对照组,差异均有统计学意义(P<0.05)。Pearson相关分析显示SLE患者外周血B细胞中miR-21和miR-155相对表达量与BMP-2 mRNA和BMPRⅡmRNA均呈正相关(r=0.292、0.365和0.481、0.279,P<0.05),SLE患者外周血B细胞中miR-21和miR-155相对表达量均与补体C3和补体C4水平呈显著负相关(r=-0.570、-0.289和-0.751、-0.308,P<0.05)。结论:miR-21和miR-155在SLE患者外周血B细胞中呈高表达,可能通过BMP信号通路异常活化而发挥作用,且与补体C3和C4呈负相关,有望作为监控SLE患者病情的参考指标。 Objective: To investigate the expressions and significances of miR-21 and miR-155 in peripheral blood B cells of patients with systemic lupus erythematosus(SLE). Methods: 93 cases of patients with SLE were divided into mild active group (n=49) and moderate and severe active group (n=44) according to SLEDAL scores. 40 healthy were selected as the control group. The expressions of miR-21, miR-155, bone morphogenetic protein-2 (BMP-2) and bone morphogenetic protein receptor II (BMPR II ) in peripheral blood B cells were detected by using real-time quantitative PCR. Results: The relative expressions of miR-21 and miR-155 in peripheral blood B ceils of SLE patients were higher than the control group (P〈0.05). The relative expressions of miR-21 and miR-155 in peripheral blood B cells in the moderate and severe active group were higher than the mild active group and the control group, and the mild active group was higher than the control group, the differences were statistically significant (P〈0.05). The relative expressions of BMP-2 mRNA and BMPR II mRNA in peripheral blood B cells of SLE patients were higher than the control group (P〈0.05). The relative expressions of BMP-2 mRNA and BMPR II mRNA in peripheral blood B cells in moderate and severe active group were higher than the mild active group and the control group, and the mild active group was higher than the control group, the differences were statistically significant (P〈0.05). Pearson correlation analysis showed, the relative expressions of miR-21 and miR-155 in peripheral blood B cells of the SLE patients were positively correlated with BMP-2 mRNA and BMPR II mRNA (r=0.292, 0.365 and 0.481, 0.279, P〈0.05), the relative expressions of miR-21 and miR-155 in peripheral blood B cells of the SLE patients were negatively correlated with the levels of complement C3 and C4 (r= -0.570, -0.289 and -0.751, -0.308, P〈0.05). Conclusion: MiR-21 and miR-155 were highly expressed in the peripheral blood B cells
