摘要
目的探讨系统性红斑狼疮(SLE)患者外周血单个核细胞(PBMC)中miRNA-155的表达水平及其对SLE预后的意义。方法收集我院2013年9月-2016年3月确诊并治疗的SLE患者61例,根据SLE疾病活动指数分为轻度组26例,中重度组35例,同期选择30例健康体检者作为对照组。应用实时荧光定量PCR技术对入选者PBMC中miRNA-155的表达水平进行检测,对比分析各组表达情况。结果与对照组相比,轻度活动组的miRNA-155表达水平无统计学差异(P>0.05);中重度活动组miRNA-155水平明显高于对照组(P<0.05)。抗dsDNA抗体、24h尿蛋白与SLE患者miRNA-155表达水平显著相关(P<0.05);与无狼疮肾组相比,狼疮肾阳性组SLE患者miRNA-155水平显著升高(P<0.05);与血液系统正常组SLE患者相比,异常组miRNA-155水平显著升高(P<0.05)。结论中重度活动SLE患者PBMC中miRNA-155表达升高;miRNA-155表达与抗dsDNA抗体及24h蛋白尿相关;miRNA-155在狼疮肾、血液系统异常中呈高表达。
Objective To investigate the Expression of minRNA-155 in peripheral blood mononuclear cells（PBMC） of systemic lupus erythematosus（SLE） patients and the the impact on prognosis. Methods 61 patients were selected from September 2013 to March 2016, who meet the diagnostic criteria of SLE from our hospital. According to SLE disease activity index 61 cases were divided into mild activity group 26 cases, moderate and severe activity group 35 cases. 30 healthy persons were selected as control group, miRNA-155 expressions in PBMC was examined by real-time PCR. Results Compared with the control group,there was no significant difference in miRNA-155 expression of mild activity group（P〉0.05）. The levels of miRNA-155 in the moderate and severe activity group were significantly higher than those in the control group（P〈0.05）. The levels of dsDNA expression and 24h were significantly correlated with the levels of miRNA-155 expression in SLE patients（P〈0.05）. Patients with lupus nephritis had higher expression levels of miRNA-155 than thoses without（P〈0.05）. Patients with abnormal blood had higher expression levels of miRNA- 155 than thoses without（P〈0.05）. Conclusion The moderate and severe activity SLE patients have higher expression of miRNA-155 in PBMC. The expression of miRNA-155 was associated with the urine of anti dsDNA and 24h proteins. MiRNA-155 was highly expressed in the abnormal renal and hematological abnormalities of lupus.
