低浓度雌二醇对骨质疏松大鼠颌骨成骨细胞增殖分化能力的影响 被引量:8

摘要 目的:研究低浓度雌二醇对骨质疏松大鼠颌骨成骨细胞增殖分化能力的影响。方法:选用3月龄健康未孕雌性SD(Sprague-Daw ley)大鼠20只,随机分为卵巢切除术组(ovx组),假手术组(sham组)。术后12w取ovx组双侧下颌骨,培养成骨细胞。采用低浓度雌二醇(10^(-10)mol/L)干预成骨细胞3d后,检测碱性磷酸酶、骨钙素的表达、PCNA(Proliferating Cell Nuclear Antigen)蛋白表达和增殖能力。结果:低浓度雌二醇明显促进了成骨细胞的增殖能力(MTT检测,雌二醇组:0.372±0.023;对照组:0.175±0.011)和PCNA蛋白表达(雌二醇组:0.495±0.023;对照组:0.176±0.012),增加了成骨细胞的碱性磷酸酶(雌二醇组:0.495±0.023;对照组:0.176±0.012)和骨钙素(雌二醇组:14.26±1.71;对照组:9.17±1.01)。结论:低浓度雌二醇增加了骨质疏松大鼠颌骨成骨细胞的增殖分化能力,可用来颌骨骨质疏松症防治的探索研究。 Objective: To investigate effect of low-concentration estradiol on proliferation and differentiation of osteoblasts from jaws of osteoporotic rats. Methods: 3-month-old non-pregnant female Sprague-Dawley(SD) rats were randomly divided into ovariectomized group(ovx group) and sham-operated group(sham group). Twelve weeks after operation, osteoblasts from bilateral mandibular from ovx group were obtained and cultured. After the osteoblasts were intervented for 3days by low-concentration estradiol(10(-10)mol/L), alkaline phosphatase, osteocalcin, PCNA protein and proliferation ability were tested. Results: The low concentration estradiol significantly promoted the proliferation(MTT assay, estradiol group:0.372±0.02, control group: 0.175 ±0.011), PCNA protein(estradiol group: 0.495 ±0.023, control group: 0.176±0.012), alkaline phosphatase(estradiol group: 0.495±0.023, control group: 0.176±0.012) and osteocalcin(estradiol group: 14.26±1.71, control group: 9.17 ±1.01). Conclusion: Low-concentration estradiol increased the proliferation and differentiation ability of osteoblasts from jaws of osteoporotic rats, and it could be used in the study of jaw osteoporosis.
