摘要
目的比较不同成分磷酸钙陶瓷材料对兔骨髓基质干细胞(BMSCs)增殖及成骨分化基因表达的影响。方法将不同相成分的磷酸钙陶瓷材料分为两组,分离并培养兔BMSCs分别与之共培养,于第1、3、7天用MTT法检测细胞增殖情况,并用FDA/PI染色后观察细胞形态。于第3、7、14天采用实时荧光定量PCR检测BMSCs的碱性磷酸酶(ALP)、骨形态发生蛋白(BMP-2)、骨唾液酸蛋白(BSP)和转录因子Osterix的mRNA表达变化。结果共培养7d后,两种材料上BMSCs均快速增殖;共培养14d时,双相磷酸钙陶瓷(BCP)组BMSCs的ALP、BMP-2、BSP和Osterix mRNA表达均明显高于羟基磷灰石(HA)组(P<0.05)。结论两种磷酸钙材料均能促进BMSCs增殖及向成骨方向分化,双相磷酸钙陶瓷这一能力强于羟基磷灰石陶瓷,预示出其更加优异的体外成骨诱导潜力。
Objective To compare the effects of different components of calcium phosphate ceramics on proliferation and osteogenic differentiation of rabbit bone marrow stromal cells (BMSCs). Methods The calcium phosphate ceramics with different phases were divided into two groups, BMSCs were isolated and cultured with ceramics. Cell proliferation was detected at day 1,3 and 7 and cell morphology was observed by FDA/PI staining. Real -time PCR was applied to detect the changes of expressions of alkaline phosphatase (ALP) , bone morphogenetic protein (BMP -2), bone sialoprotein (BSP) and transcription factor Osterix at day 3,7 and 14. Results After 7 days of co - culture, BMSCs on two kinds of materials had good morphology and wellproliferation. At 14d of co - culture,the mRNA expression of ALP, BMP -2, BSP and Osterixinbiphasic calcium phosphate ceramics group were significantly higher than those in HA group ( P 〈 0.05 ). Conclusion Both BCP and HA could promote the proliferation and osteogenic differentiation of BMSCs. The osteogenic differentiation ability of BCP is superior to that of HA to indicate its osteoinductive activity in vitro.
