Genetic Characterization of Nucleoprotein of Measles Virus Strains Circulating among Children in Hangzhou and Its Peripheral Areas,2012-2013

摘要 目的探讨2012~2013年杭州及周边地区儿童麻疹病毒(Measles Virus,MV)流行株核蛋白的基因特征。方法用逆转录-聚合酶链反应(RT-PCR)检测麻疹患者的咽拭子标本,对部分PCR扩增阳性的片段进行基因测序、并绘制系统进化树进行同源性分析。结果从杭州市儿童医院2012~2013年80例儿童麻疹患者的咽拭子中检测到29例MV株,经基因测序及系统进化树分析后均为H1a基因型,与该型参考株china93-2﹑china93-4核苷酸同源性为97.7%~98.9%;2012年基因型组内遗传距离(0.007)最小,2012年和2013年基因型之间的组间遗传距离(0.013)最小,而2012年、2013年基因型与A型(疫苗株)的组间遗传距离均较大,分别为0.085、0.087。结论 2012~2013年杭州地区儿童MV的优势亚型为H1a亚型,与中国其他地区报告的流行株一致。2012年与2013年的MV基因比较遗传变异较小,而与疫苗株比较遗传变异较大。 Objective To discuss genetic characterization of measles virus (MV) circulating among children in Hangzhou and its peripheral areas, 2012-2013. Methods The reverse transcription-polymerase chain reaction (RT-PCR) was used to detect throat swab specimens from suspected measles cases, and to analyze the sequence of gene segments of part PCR products. Results 29 MV strains were isolated from 80 throat swab specimens of suspected measles cases patients from Hangzhou Children's Hospital in 2012-2013. By sequencing gene segments of PCR products, all strains were identified as Hla genotypes. The nucleotide homology between Hl a genotype and the reference strain (china 93-2,china 93-4) ranged from 97.7% to 98.9%. The p-distance (0. 007) within genotypes in 2012 was the smallest. The p-distance (0. 013) between genotypes in 2012 and 2013 was the smallest. The significant distances were found between genetype A (vaccine strain ) virus and genotypes in 2012 or 2013 , which were 0. 085 and 0. 087, respectively. Conclusions HI a genotype is the ascendant subgenotype of the local epidemic MV strains in Hangzhou, which accorded with reported epidemic strains of other areas in China. It had the less nucleotide variation between genotypes in 2012 and 2013, but the higher variation between genetypes in 2012 and the vaccine strain.
