摘要
目的:探讨益气化瘀方对糖尿病溃疡大鼠血管再生及溃疡创面愈合能力的影响。方法:SPF级大鼠90只,采用随机数字表法将其分为3组,每组30只,益气化瘀组予益气化瘀方灌胃,模型组、空白组均予生理盐水灌胃;采用STZ和高糖高脂饮食制造糖尿病大鼠模型;每只大鼠手术制造一边长2 cm的正方形皮肤创面。术后3、7、14 d每次随机在各组中取10只大鼠,观察大鼠创面愈合率、取溃疡标本Western-blot检测标本VEGF、VEGFR,免疫组化观察微血管密度。结果:益气化瘀组和模型组术后3、7、14 d创面愈合率明显低于空白组(P<0.05)。益气化瘀组术后3、7、14 d创面愈合率高于模型组(P<0.05)。益气化瘀组和模型组术后3 d、7 d VEGF、VEGFR光密度值明显低于空白组(P<0.05)。术后14 d,益气化瘀组VEGF、VEGFR光密度值高于空白组,差异有统计学意义(P<0.05)。益气化瘀组术后3、7、14 d VEGF、VEGFR光密度值均高于模型组(P<0.05)。益气化瘀组术后3、7 d微血管密度低于空白组(P<0.05)。术后14 d,益气化瘀组微血管密度高于空白组(P<0.05)。益气化瘀组术后3、7、14 d微血管密度均高于模型组(P<0.05)。结论:益气化瘀方能改善糖尿病大鼠溃疡局部VEGF、VEGFR分子的分泌,促进溃疡面的愈合。
Objective： To study the influence of Yiqi Huayu Prescription （YQHYP, 益气化瘀方） on vascular regeneration and ulcer wound healing ability in diabetic rats. Methods： The 90 SPF rats were randomly divided into 3 groups and each group 30. YQHYP group was given Yiqi Huayu Prescription （5 g/day, 2 mL）, model group and control group were treated with normal saline lavage （2 mL）. Diabetic rats were made by STZ and with high sugar and fat diet. Each rat was made a 2 cm long square skin wounds. At 3 days, 7 days, 14 days after the surgery, 10 rats were taken from each group. Wound healing rate was observed of ulcer specimen. The level of VEGF and VEGFR were tested by Western blot, and microvascular density was tested by immunohistochemistry. Results： The wound healing rate of YQHYP group and model group were significantly lower than the control group at 3 d, 7 d, 14 d after the surgery （P〈0.05）, The wound healing rate of YQHYP group was higher than the model group at 3 d, 7 d, 14 d after the surgery （P〈0.05）. The VEGF and VEGFR optical density value of YQHYP group and model group was lower than the control group significantly at 3 d, 7 d after the surgery （P〈0.05）, while that in YQHYP group was significantly higher than the control group at 14 d after the surgery and the difference was statistically significant （P〈0.05）. The VEGF and VEGFR optical density values of YQHYP group were higher than the model group at 3 d, 7 d, 14 d after the surgery （P〈0.05）. Microvascular density in YQHYP group was lower than the control group at 3 d, 7 d after the surgery （P〈0.05）. Microvascular density in Yiqihuayu Prescription group was higher than the control group at 14 d dafter the surgery （P〈0.05）. The Microvaseular density of YQHYP group was higher than the model group at 3 d, 7 d, 14 d after the surgery （P〈0.05）. Conclusions： YQI-IYP can improve the secretion of VEGF and VEGFR molecules of ulcer local in diabetes rats, and promote healing of the ulcer surface.
