胚胎中脑神经干细胞移植治疗大鼠帕金森病模型 被引量:5

摘要 目的研究胚胎中脑神经干细胞(mNSCs)移植对帕金森病(PD)模型大鼠的治疗作用。方法分离培养大鼠胚胎mNSCs并进行EGFP基因修饰。前脑内侧束和纹状体立体定向注射6-羟多巴胺建立PD大鼠模型。将EGFP基因修饰mN SCs定向移植到PD大鼠纹状体区。免疫荧光组织化学鉴定移植细胞的存活、迁移和分化。行为学实验观察治疗效果。结果 EGFP基因修饰胚胎mN SCs移植能分化为多巴胺神经元和小胶质细胞。移植后行为学实验发现可改善运动功能。结论移植EGFP基因修饰胚胎mNSCs可改善PD大鼠的运动障碍。
