电刺激对缺氧缺血性脑损伤新生大鼠脑组织bFGF的影响 被引量:6

Effect of electrical stimulation of b FGF in brain tissue of neonatal rats with hypoxic ischemic brain damage

摘要 目的探讨电刺激对缺氧缺血性脑损伤新生大鼠脑组织碱性成纤维细胞生长因子(b FGF)的影响。方法选取60只7日龄清洁级SD雄性大鼠随机分为电刺激组、模型组以及假手术组,制作大鼠缺氧缺血性脑损伤模型。电刺激组为大鼠术后12h进行电刺激治疗,模型组和假手术组不给予刺激。实验结束后各取5只大鼠处死,以免疫组化技术检测b FGF阳性细胞数,以RT-PCR检测b FGF基因水平表达。结果电刺激组、模型组和假手术组第3、7、14、21d在海马齿状回区Brd U阳性细胞数分别为(24.50±5.07、53.13±5.44、47.88±5.25、19.75±4.53)、(13.00±3.74、23.75±4.17、19.00±3.93、14.23±3.92)、(13.25±3.84、18.50±3.59、20.25±4.40、14.25±3.81),模型组和假手术组不同时间段Brd U阳性细胞数差异无统计学意义(P>0.05),电刺激组高于模型组和假手术组(P<0.05)。电刺激组、模型组和假手术组第3、7、14、21d大鼠b FGF m RNA分别为(0.36±0.12、0.75±0.05、0.72±0.13、0.35±0.10)、(0.24±0.04、0.41±0.07、0.32±0.06、0.25±0.05)、(0.24±0.06、0.25±0.05、0.28±0.04、0.25±0.04),电刺激组大鼠不同时间段b FGF m RNA表达显著高于模型组和假手术组(P<0.05),模型组和假手术组差异无统计学意义(P>0.05)。结论缺氧缺血性脑损伤新生大鼠通过电刺激后脑组织b FGF阳性细胞数目增多,能够加快其脑部神经元修复。 Objective To investigate the effect of electrical stimulation on brain tissue bFGF in ischemic brain injury in neonatal rats. Methods The data of 60 healthy SD rats with 7 day-old were randomly divided into electrical stimulation treatment group, the model group and the sham operation group, the ischemic brain injury model were made in rats. Electrical stimulation treatment group were treated by the electric stimulation 12 h after operation, the model group and the sham operation group were not treated by the electric stimulation. After the end of the experiment, 5 rats were sacrificed and the number of bFGF positive cells was detected by immunohistochemical technique. The expression of bFGF gene was detected by RT-PCR technique. Results The number of BrdU positive cells in the dentate gyrus region of the electrical stimulation treatment group , the model group and the sham operation group were (24.50±5.07,53.13±5.44,47.88±5.25,19.75±4.53), (13.00±3.74,23.75±4.17,19.00±3.93,14.23±3.92) and( 13.25±3.84,18.50±3.59,20.25±4.40,14.25±3.81 )after 3 d, 7 d, 14 d and 21 d, respectively. The number of BrdU positive cells in the dentate gyrus region between the model group and the sham operation group were not significantly different (P〉0.05). The BrdU positive cells in the hippocampus of rats in different time periods were significantly higher than those in the model group and sham operation group (P〈O.05). The expression of mRNA bFGF in the dentate gyrus region of the electrical stimulation treatment group, the model group and the sham operation group were (0.36±0.12,0.75±0.05,0.72±0.13,0.35±0.10), (0.24±0.04,0.41 ±0.07,0.32±0.06,0.25±0.05) and (0.24±0.06,0.25± 0.05,0.28±0.04,0.25±0.04)after 3 , 7,14 .and 21 d, respectively. The expression of mRNA bFGF in the electrical stimulation treatment group was significantly higher than that in model group and sham operation group (P〈0.05); the expression of mRNA bFGF was not significantly different in model group a
