摘要
目的:观察淫羊藿苷对去势大鼠血清E_2水平及骨组织ERB基因表达的影响。方法:将72只雌性SD大鼠建立去卵巢骨质疏松症病理模型,随机分为淫羊藿苷高剂量组(A)、淫羊藿苷低剂量组(B)、壮骨止痛方中剂量组(C)、模型组(D)、戌酸雌乙醇对照组(E),另选取12只雌性SD大鼠为假手术组(F)。各组分别灌胃给药13周。每组随机选取8只实验大鼠经腹主动脉采血,凝固后离心采用酶联免疫吸附法(ELISA)测定检测各组血清E_2浓度,并提取股骨组织mRNA样本进行PCR反应,检测ERβ基因的表达。结果:各给药组血清E_2水平上升明显,ERβ基因表达回调明显,A、C、E、F组较D组血清E_2水平上升、ERB基因表达上调差异均有统计学意义(P<0.05,P<0.01);B组上述指标与F组比较,差异有统计学意义(P<0.05,P<0.01)。结论:淫羊藿苷可能通过改善去势大鼠血清E_2水平,同时上调去势大鼠骨组织ERβmRNA的表达,使ERβ合成增加,从而提高ER的生物效应,达到抗绝经后骨质疏松的作用。
Objective:To investigate the effects of icariin on serum estradiol(E_2) level and estrogen receptor β(ERβ) gene expression in bone tissue in ovariectomized rats.Methods:A total of 72 female Sprague- Dawley(SD)rats were selected and ovariectomized to establish the pathological model of osteoporosis,and randomly divided into high- dose icariin group(group A),low- dose icariin group(group B) group with a medium dose of bone- strengthening and pain- relieving prescription(group C),model group(group D),and estradiol valerate control group(group E);another 12 female SD rats were selected as the sham- operation group(group F);all the drugs were administered by gavage for 13 weeks.Eight rats were randomly selected in each group,and blood samples were collected from the abdominal aorta and were centrifuged after coagulation.The enzyme- linked immunosorbent assay was applied to measure serum E_2 level;mRNA samples were collected from femoral tissue and polymerase chain reaction was performed to measure ERβ gene expression.Results:All the administration groups showed a significant increase in serum E_2 level and a significant reduction in ERβ gene expression,and compared with group D groups A,C,E,and F had a significant increase in serum E_2 level and significantly upregulated expression of ERβ gene(P〈0.05,P〈0.01);the indicators above showed a significant difference between group B and group F(P〈0.05,P〈0.01).Conclusion:By improving serum E_2 level in ovariectomized rats,upregulating the mRNA expression of ERβ in bone tissue,and increasing the synthesis of ERβ,icariin can improve the biological effect of ER and achieve the anti- postmenopausal osteoporosis effect.
